牛磺酸螯合钙的制备、纯化、表征及透皮吸收的研究

【摘要】 钙是人体中含量最多的矿物质元素,不仅可以调节人体的生命活动及代谢,对维持人的皮肤健康也发挥重要的作用。人体皮肤表层的钙离子浓度呈梯形分布,这种分布对调节角质细胞增殖分化,维持表皮的屏障功能发挥重要的作用,对于损伤的皮肤表层,可以促进其自我修复;一些钙制剂可以缓解炎症引发的皮肤瘙痒。目前对于螯合钙的研究较多,但对于牛磺酸螯合钙的研究极少,将螯合钙与皮肤补钙结合的研究也未见报道。本课题主要对一种新型补钙剂——牛磺酸螯合钙展开研究,内容主要包括牛磺酸螯合钙的制备、纯化、表征及透皮吸收四部分。在牛磺酸螯合钙的制备部分,对氧化钙和氯化钙两种钙源制备牛磺酸螯合钙的工艺进行条件优化,得到氧化钙为钙源的螯合钙的最优条件为牛磺酸浓度0.8 mol/L,氧化钙添加量0.4mol/L,在90℃恒温水浴锅中磁力搅拌反应30min,反应结束后抽滤除去未反应的氧化钙固体即得到螯合钙溶液;以氯化钙作为钙源制备牛磺酸螯合钙的最优工艺条件为:牛磺酸0.8 mol/L,氯化钙0.4 mol/L,70 ℃旋转蒸发至干,再用乙醇洗涤除去未反应的氯化钙,干燥得到牛磺酸螯合钙固体。在牛磺酸螯合钙的纯化部分,尝试了乙醇沉淀法、离子交换树脂法和溶解度差异法三种纯化方法,并对其纯化效果进行分析比较,结果表明,以氯化钙为钙源制备螯合钙结合乙醇洗涤除杂纯化效果最好。本研究主要采取的表征手段有质谱、红外光谱检测、核磁共振氢谱等,确定了牛磺酸螯合钙的相对分子量为288,推测其分子式为C4H12N2O6S2Ca,得出牛磺酸螯合钙是由两个牛磺酸与一个钙离子结合而成,结合方式为两个牛磺酸的磺酸基和氨基共同作用于中心钙离子,形成两个相连六元环状结构。最后,比较了牛磺酸螯合钙与乳酸钙和氯化钙的透皮吸收率,得出结论:牛磺酸螯合钙的透皮吸收率明显高于乳酸钙和氯化钙,表明牛磺酸螯合钙易吸收,生物利用率高,有望添加到化妆品及一些皮肤病药物中对皮肤进行补钙。

【Abstract】 Calcium is the most abundant mineral element in the human body,Calcium plays an important role in the life activity of human,which wildly existed in skin.The calcium’s concentration in trapezoidal distribution in the surface of human skin,and this distribution way in skin allows it easier to regulate keratinocytes proliferation and differentiation,maintain the barrier function of epidermis,and enhance the self-repair ability of damaged skin.Calcium preparations has curative effect on pruritus senilis caused by inflammation.Although there are many researches conducted on chelating calcium,few of them are related to taurine chelating calcium,especially on the relationship between chelating calcium and skin calcium knot.This project was research on preparation,purification,characterization and transdermal absorption of taurine chelating calcium.In the part of preparation,the chelating process was optimized with calcium oxide and calcium chloride was used as calcium source.According to the results,0.8 mol/L taurine combined with 0.4 mol/L calcium oxide at 90 ℃ for 30 min was the optimal reaction condition,and 0.8mol/L taurine combined with 0.4 mol/L calcium chloride at 70 ℃ was the optimal reaction condition.Purification effect was evaluated by ethanol precipitation,ion exchange resin and different solubility.the results showed that the preparation of chelating calcium with calcium chloride as the calcium source,washing with ethanol can get the product with highest purity.The structure of taurine chelating calcium was characterized by Mass Spectrometry,Infrared Spectroscopy,Nuclear Magnetic Resonance.The relative molecular mass of taurine chelating calcium was determined as 288,and it’s molecular formula is C4H12N2O6S2Ca,And we got that the taurine chelating calcium is made up of two taurine and one calcium ion with the structure of two six-member rings connected each other,because of sulfo group and amino of taurine interacting with the calcium ion.Finally,transdermal absorption rate was compared between taurine chelating calcium,calcium lactate and calcium chloride,taurine chelating calcium showed obviously higher rate than that of calcium lactate and calcium chloride.That taurine chelating calcium possesses the advantages of absorptivity and bioavailability.It is expected to be added to cosmetics and some drugs for skin diseases to supplement calcium for skin.
