化学杀菌剂复合处理对植物乳杆菌烈性噬菌体的影响

【摘要】 工业生产中常使用化学杀菌剂对环境及设备进行消毒杀菌。但是,单一化学杀菌剂对噬菌体灭活影响不大。评价多种化学剂复合使用对噬菌体的灭活效果,可为工业上建立噬菌体防御体系提供有效依据。本实验将以乳杆菌烈性噬菌体P1及P2为研究对象,研究不同浓度的乙醇与不同浓度异丙醇、次氯酸钠、过氧乙酸等化学试剂复合处理10 min对其的灭活效果。处理方法包括:乙醇溶液与其他化学杀菌剂直接混合灭活10 min;乙醇溶液灭活5 min后,加入其他化学杀菌剂灭活5 min;其他化学杀菌剂灭活5 min后,加入乙醇溶液处理5 min。实验结果表明:(1)采用复合灭活方法可有效灭活噬菌体P1、P2。可在满足商业杀菌消毒要求的同时,降低成本,保证相关产品的稳定性。(2)100%异丙醇与100%乙醇直接混合10 min,可以完全灭活噬菌体P1。先用100%乙醇处理5 min再用100%异丙醇处理5 min或先用100%异丙醇处理5 min再用100%乙醇处理5 min,可使噬菌体P2完全失活。(3)800ppm次氯酸钠处理5 min后加入20%乙醇灭活5 min或75%乙醇处理5min后再加入400ppm次氯酸钠处理5 min或800ppm次氯酸钠处理5 min后加入75%乙醇均可使噬菌体P1完全失活。200ppm次氯酸钠处理5 min后再用75%乙醇灭活5min,或200ppm次氯酸钠处理5 min后加入100%乙醇,即可完全灭活噬菌体P2。(4)乙醇与过氧乙酸复配并不能使噬菌体P1、P2完全失活。先用100%乙醇处理5 min后加入0.45%过氧乙酸,可使噬菌体P1降低3.16个对数级。100%乙醇处理5 min后加入0.45%过氧乙酸灭活5 min,可使噬菌体P2下降3.84个对数级。

【Abstract】 Biocides are often used to disinfect the environment and equipment in industrial production.However,single chemical biocide expressed limited effect on phage inactivation.Evaluation of the combined inactivate effect of multiple biocides on phage may provide effective basis for the establishment of phage defense systems in industry.In this study,the combined inactivate effect of ethanol and other biocides(isopropanol,sodium hypochlorite and peracetic acid)with different concentrations on Lactobacillus bacteriophage P1 and P2 were evaluated.Moreover,three compound methods were used,including directly mixing the biocides and treated for 10 min;treated by ethanol solution for 5 min followed by other biocide for 5 min;treated by other biocide for 5 min followed by ethanol solution for 5 min.The results were listed below.(1)Combined biocides can inactivate Phage P1 and Phage P2 effectively.It can meet the requirements of commercial sterilization,reduce the cost,and guarantee the stability of related product.(2)Lactobacillus phage P1 can be totally inactivated by the mixture of 100% isopropyl alcohol and 100% ethanol for 10 min.Lactobacillus phage P2 can be totally activated by 100 % ethanol treated for 5 min followed by 5 min 100% isopropanol treatment,vice versa.(3)800 ppm sodium hypochlorite for 5 min followed by 20% ethanol treated for 5 min,or 75% ethanol for 5 min followed by 400 ppm sodium hypochlorite,or 800 ppm sodium hypochlorite treated for 5min followed by 5 min 75% ethanol treatment can inactivate phage P1 completely.Lactobacillus phage P2 can be totally inactivated by either 200 ppm sodium hypochlorite treated for 5 min and then inactivated 5 min by 75% ethanol or 200 ppm sodium hypochlorite followed by 100% ethanol.(4)The combination of ethanol and peracetic acid can not inactivated these two phages completely.For phage P1,100% ethanol followed by 0.45% peracetic acid only resulted in 3.16 log reduction.For phage P2,it only caused 3.84 log reductions.
