Sema4D和CXCR4在肺癌组织中表达及与血管生成、转移的关系

【摘要】 目的:研究Sema4D、CXCR4在肺癌组织中的表达,探讨其与肺癌患者的预后及临床病理因素的关系。方法:采用免疫组化方法检测95例非小细胞肺癌组织标本、20例正常肺组织标本中的Sema4D、CXCR4表达水平,分析其与肺癌临床病理因素之间的关系。采用免疫组化方法,检测95例非小细胞肺癌组织、20例正常肺组织标本中CXCR4的表达及微血管密度(MVD)的值,并分析CXCR4的表达与MVD之间的关系,以及与肺癌临床病理因素之间的关系。采用RT-PCR方法检测95例肺癌组织、20例正常肺组织中CXCR4表达水平,与免疫组化方法检测结果进行对比分析。结果:Sema4D、CXCR4在非小细胞肺癌组织中的表达明显高于正常肺组织(P<0.05),其表达水平与患者性别、年龄、病理类型、分化程度无关(P>0.05),与淋巴结转移、TNM分期相关(P<0.05)。非小细胞肺癌组织中Sema4D与CXCR4的表达无明显相关性(P>0.05)。非小细胞肺癌组织中CXCR4的表达以及MVD值明显高于正常肺组织(P<0.01),CXCR4的表达水平与MVD值呈正相关(P<0.01)。免疫组化SP法和RT-PCR法检测CXCR4在非小细胞肺癌组织中的表达水平,两种方法检测结果基本一致。结论:临床上检测非小细胞肺癌患者肿瘤组织中Sema4D和CXCR4的表达,在研究肺癌的发生、发展和判断预后等方面有着积极的指导意义,有可能成为治疗非小细胞肺癌的新靶点。
【Abstract】 Purpose:To investigate the expression of Sema4D,CXCR4 in lung cancer and to explore its relationship with prognosis and clinicopathological parameters of patients with lung cancer.Method:Immunohistochemistry was used to detect Sema4D,CXCR4 expression in 95 cases of non-small cell lung cancer tissues,and 20 cases of normal lung tissue specimens,analyzing the relationship between the clinical and pathological factors of lung cancer.Immunohistochemistry was used to detect expression of CXCR4 and microvessel density(MVD)value to analyze the relationship between the expression of CXCR4 and the MVD,as well as the relationship between lung cancer the relationship and pathological factors.RT-PCR was used to detect levels of CXCR4,comparing the results of immunohistochemistry test.Results:The expression Sema4D and CXCR4 in non-small cell lung cancer tissues was significantly higher than that in normal lung tissues(P<0.05),and its expression has nothing to do with gender,age,pathological type,differentiation of patients(P>0.05),but was associated with lymph node metastasis,TNM staging(P<0.05).There was no significant correlation between expression of CXCR4 and Sema4D in non-small cell lung cancer(P>0.05).The expression of CXCR4 and MVD value were significantly higher in Non-small cell lung cancer tissues than in normal lung tissues(P<0.01),the expression level of CXCR4 and MVD value were positively correlated(P<0.01).The results of CXCR4 expression in non-small cell lung cancer detected by immunohistochemical method and RT-PCR method were consistent.Conclusion:The clinical detection of Sema4D and CXCR4 expressions in tumor tissue from patients with non small cell lung cancer has positive significance in the study of lung cancer occurrence,development and prognosis,etc.,and may become a new target for treatment of non-small cell lung cancer.
