两种低聚果糖理化特性及对益生菌作用的研究

Study on the Physical and Chemical Properties of Two Kinds of Fructo-oligosaccharides and Their Effects on Probiotics

【摘要】 目前,根据生产原料和工艺的不同,低聚果糖可以分为两种:菊粉来源的低聚果糖(简称"菊粉低聚果糖")和蔗糖来源的低聚果糖(简称"蔗糖低聚果糖")。本研究测定了两种低聚果糖的水分活度、美拉德反应和热稳定性,比较两种低聚果糖在理化特性上的异同,结果发现:(1)菊粉低聚果糖的水分活度为0.1625,蔗糖低聚果糖的水分活度为0.1738;(2)两种低聚果糖产品发生轻微的美拉德反应;(3)菊粉低聚果糖的热变性温度是64.3℃,蔗糖低聚果糖的热变性温度是60.5℃。有研究表明,两种低聚果糖均能调节肠道菌群,促进肠道内双歧杆菌等益生菌的增殖。本研究通过体外实验和体内实验,评价两种低聚果糖对益生菌的增殖效果。体外实验:分别以菊粉低聚果糖和蔗糖低聚果糖为碳源,测定益生菌的生长曲线,比较两种低聚果糖对益生菌的体外增殖作用是否等同。体内实验:选取120只昆明雄性小鼠,分成10组,每组12只,灌胃不同剂量的益生菌菌粉(两种双歧杆菌菌粉)和低聚果糖,断颈致死,取肠道内容物,利用传统培养法、普通PCR和实时荧光定量PCR,研究肠道内益生菌(双歧杆菌和乳杆菌)的增殖情况。结果表明:(1)对小鼠灌胃两种低聚果糖,对于小鼠的正常生长和饮食无不良影响;(2)两种低聚果糖对双歧杆菌和乳杆菌的生长增殖均表现出促进作用;(3)两种双歧杆菌菌粉对双歧杆菌和乳杆菌的生长增殖均表现出促进作用;(4)从体外实验部分生长曲线来看,菊粉低聚果糖对两种双歧杆菌的促生长作用整体上优于蔗糖低聚果糖;从体内实验部分来看,菊粉低聚果糖对双歧杆菌和乳杆菌的促生长作用也优于蔗糖低聚果糖。结果表明:整体上,菊粉来源的低聚果糖对益生菌的促生长作用优于蔗糖来源的低聚果糖。

【Abstract】 At present,according to the differences of the raw materials and the producing process,fructo-oligosaccharides could be divided into two kinds:FOSinulin and FOSsucrose.This study measured the water activity,Maillard reaction and thermal stability of two kinds of fructo-oligosaccharides to compare the differences of the physicochemical properties of FOSinulin and FOSsucrose.The results are as follows:(1)The water activity of FOSinulin is 0.1625,and the water activity of FOSsucrose is 0.1738;(2)Both two kinds of fructo-oligosaccharide products could have mild Maillard reaction;(3)The thermal denaturation temperature of FOSinulin is 64.3℃,and the thermal denaturation temperature of FOSsucrose is 60.5℃.Some studies have indicated that two kinds of fructo-oligosaccharides could regulate the intestinal flora,and promote the proliferation of probiotics in the intestinal tract.The effect of two kinds of fructo-oligosaccharides on the proliferation of probiotics was evaluated in vitro and in vivo experiments.The growth curve of probiotics with FOSinulin and FOSsucrose as carbon source in the culture medium respectively were got to see if two kinds of fructo-oligosaccharides had the same proliferation in vitro on probiotics in vitro experiment.In vivo experiment:120 male mice in Kunming were divided into 10 groups,12 rats in each group,and we would feed these mice the different doses of probiotic bacteria powder(two kinds of bifidobacterium bacteria powder)and fructo-oligosaccharides;The intestinal contents would be got,and the traditional culture method,PCR and real-time fluorescent quantitative PCR were used to study the proliferation of intestinal probiotics(Bifidobacterium and Lactobacillus).The results show:(1)Two kinds of fructo-oligosaccharides have no adverse effect on the normal growth and diet of the mice;(2)Both two kinds of fructo-oligosaccharides could promote the proliferation of bifidobacterium and lactobacillus;(3)Two kinds of bifidobacterium bacteria powder could promote the proliferation of bifidobacterium and lactobacillus;(4)From the growth curve in vitro,the FOSinulin has a better effect on two kinds of bifidobacteriums compared with the FOSsucrose in the overall;And from experiments in vivo,the FOSinulin has a better effect on the growth of bifidobacteria and lactobacillus compared with FOSsucrose.In this study,it turns out that the FOSinulin has a better effect on the growth of probiotics compared with FOSsucrose.
