深层液体发酵冬虫夏草菌粉的研究

【摘要】 冬虫夏草(Cordyceps sinensis),又名中华虫草、夏草冬虫,简称虫草,是中国传统的名贵中药材。虫草多糖、虫草酸和虫草素是冬虫夏草的主要活性物质。但由于自然界中野生虫草越来越少,而且价格很高,目前偏向于工厂发酵法生产。经多年的临床使用表明,冬虫夏草人工菌丝制品的药理作用与天然冬虫夏草几乎一致,个别有益指标甚至高于天然冬虫夏草,但其价格要远远低于天然虫草。因此通过冬虫夏草深层液态发酵获得菌丝体和代谢产物非常有实际价值。本文以天然冬虫夏草分离培养出的菌株为出发菌种,进行了深层液体发酵生产冬虫夏草菌粉的相关研究工作,通过改变发酵培养条件考察了其在不同的生理环境条件下,菌丝体的形态、结构、质量指标等性能参数的变化趋势,为其在规模化生产中的工艺优化提供理论参考。重点研究了培养基中各种碳氮源对菌丝生长过程中的影响,包括不同厂家的鱼胨、肉胨、酵母膏、葡萄糖、液化酶等对产品产量与质量的影响。结果表明,发酵配方中的C/N对生产的产量与质量密切相关。同时也发现,增加碳源可以提高产物甘露醇的合成,但不能保证生产中产量的稳定。在培养基优化的基础上,又对菌丝生长的环境进行了优化实验。结果表明,发酵液pH不是影响甘露醇含量的决定性因素,却可能会对菌粉腺苷、残渣等质量指标有影响;研究还发现,控制九天左右的发酵周期,可以有效地防止甘露醇的下降,并能能提高单罐放罐产量;提高罐压并不能很好地提高甘露醇的含量,对菌丝干重的提高也不是很稳定。论文还重点研究了发酵液菌丝生长过程中的补料方式,发现消毒的形式以及补料的次数都会影响生产的产量与质量。生产补料采用葡萄糖分消工艺控制,产量可提高15%左右,并且有利于在提炼过程中对发酵冬虫夏草菌粉颜色的控制。发酵生产上多批次补料对产量的提高有一定的帮助,但要在发酵中后期合成甘露醇时有效保证供养需求,才能不影响质量。
【Abstract】 Cordyceps sinensis is a traditional Chinese herbal medicine.Cordyceps sinensis by clavicipitaceae insect genus Hirsutella sinensis parasitic Hepialus armoricanus larvae in Alpine Meadow Soils.Cordyceps polysaccharide,Cordyceps acid and cordycepin is the main active substance of Cordyceps sinensis.But due to the less wild Cordyceps and nature,and the price is very high,the current bias in factory production by fermentation.After many years of clinical use showed that the pharmacological effects of Cordyceps products and natural Cordyceps is almost the same,some useful indicators even higher than that of natural Cordyceps sinensis,but its price to far lower than that of the natural Cordyceps.Therefore by Cordyceps sinensis in submerged fermentation obtained mycelium and metabolite very practical value.In this paper,natural Cordyceps isolated strains of bacteria,in the production process of Cordyceps deep liquid fermentation,by changing the fermentation to investigate the physiological environment in different conditions,conditions of mycelia morphology,structure,specific trends and quality indexes of performance parameters,to provide a theoretical reference for the in production process optimization.Focus on the research medium various carbon and nitrogen sources on mycelial growth process in the different effects.The effects of different manufacturers,fish peptone,meat peptone,yeast extract,glucose,enzymatic liquefaction on Yield and quality in production.Fermentation formula in the C/N ratio on production yield and quality is closely linked.To increase the carbon source can improve product mannitol synthesis,but can not guarantee the production yield stability.Based on the optimization of the medium,of mycelial growth environment of experimental optimization.The decisive factors of the fermentation broth pH did not affect the content of mannitol,but may effects to adenosine bacteria powder,residue and other quality indicators;fermentation period such as control in about nine days,can effectively prevent mannitol decreased,and as far as possible,to increase the yield,improve the tank pressure and not very good increase the mannitol content and on mycelial dry weight increase is not very stable.Focuses on the liquid fermentation mycelia growth in the process of feeding mode,a disinfect form and fill the number of material will effect on the yield and quality.Production fed with glucose consumer process control,yield was significantly increased by 15%,and to refine the control of fermentation Cordyceps fungi powder color.Fermentation batch feeding on Yield improvement of some help,but to in the fermentation,post synthesis of mannitol to ensure effective support demand,in order not to affect the quality.
