安康地方中蜂蜜理化成分及抗氧化活性研究

【摘要】 安康地处中华蜜蜂养殖保护区,素有养蜂传统,有一定规模。所产中蜂蜜大都为多花蜜,产量不高,未形成产业规模。各县区中蜂蜜的颜色从浅琥珀色到深琥珀色不等,有的甚至深红发黑,结晶难易程度不一。本文对安康地方中蜂蜜的理化成分和硒元素含量、游离氨基酸、抗氧化活性等几个方面进行了分析和研究。1、安康地方中蜂蜜的理化指标测定。测定的内容包括安康地方中蜂蜜样品的灰分、水分含量、pH、电导率值、果糖、葡萄糖、蔗糖、总酸、蛋白质含量、游离酸含量、内酯酸含量、淀粉酶活性。结果显示所测安康地方中蜂蜜理化指标符合我国国家标准(GB18796-2005)和国际食品法典委员会(Codex Alimentarius Commission简称CAC)的标准。2、采用氢化物—原子荧光法测定硒含量,通过对安康地方中蜂蜜中硒含量的测定来探究安康地方中蜂蜜硒含量与产地的关系。实验结果表明:安康中蜂蜜硒含量在1.483 ug/kg到43.326 ug/kg之间,中蜂蜜硒含量的高低与土壤含硒水平有关,从而为安康地方天然含硒蜂蜜特色产品开发提供依据。3、测定了安康地方中蜂蜜中氨基酸的组成和含量,实验选用的是反相高相液相色谱法。分析研究发现,安康地方中蜂蜜中的游离氨基酸数量较多且种类较齐全,总游离氨基酸含量在0.123~2.025 mg/g之间,各种氨基酸的含量占比未见明显规律。不同产地的中蜂蜜中游离氨基酸组成和含量差异显著。4、以安康地方中蜂蜜为研究对象,对其酚酸含量、DPPH自由基清除能力、还原力进行一系列实验检测。实验结果表明,安康地方中蜂蜜均具有抗氧化性,且抗氧化程度不同。其中DPPH自由基清除、还原力都与蜂蜜中酚酸含量呈正比关系,且两者能力的强弱都成剂量关系。酚酸含量越高的安康地方中蜂蜜,其DPPH自由基清除能力越强,还原力也越强,进而具有较好的抗氧化能力。
【Abstract】 Ankang is located in the Chinese bee breeding area, it known as the traditional apiculture, has been got a certain scale. Chinese honey of Ankang is a kind of healthy place Multi nectar. Most of the honey produced Different floral origin, the output is not high, not formed industrial scale. The color of honey in different areas from the light amber to deep amber range, and some even dark red, Crystallization degree is variant. In this paper, the physical and chemical composition of honey, selenium content, free amino acid, antioxidant activity and so on were analyzed and studied.1. Physicochemical characterization:including the moisture content, ash content, conductivity value, pH, free acid content, lactic acid content, total acid, protein content, amylase activity, fructose, glucose, sucrose content of the determination. The results showed that the physical and chemical indexes of honey in Ankang place accorded with the national standard of China and the Codex Alimentarius Commission (CAC) and the national standard of China.2. Selenium content is determined by Selenium content is determined by hydride atomic fluorescence method. The relationship between selenium content and origin in honey was studied. The results showed that the content of selenium in honey was 1.483 ug/kg to 43.326 ug/kg, and the content of selenium in honey was related to the content of selenium in soil, which provided the basis for the development of natural selenium-containing honey products in Ankang.3.The composition and content of free amino acids in honey were analyzed by reversed phase high performance liquid chromatography (HPLC). The results showed that the content of free amino acids in honey was large and full range, the content of total free amino acids was 0.123-2.025 mg/g, the content of various amino acids accounted for no obvious rule. There were significant differences in the composition and content of free amino acids in honey from different regions.4. Take Ankang honey as the research object, the phenolic acid content, DPPH free radical scavenging ability and reducing power were tested in a series of experiments. The results showed that the honey in Ankang has antioxidant activity, especially different levels of antioxidant activity. The DPPH free radical scavenging and reducing power were in direct proportion to the content of phenolic acid in honey, the most important was the strength of both had a dose-dependent relationship. It is to say, the higher the content of phenolic acids, the stronger the DPPH free radical scavenging ability, the stronger the reduction power, and the stronger the antioxidant capacity.
