苯环喹溴铵对脂多糖诱导的人鼻黏膜上皮细胞表达MUC5AC的影响

【摘要】 目的:探讨苯环喹溴铵(bencycloquidium bromide,BCQB)对脂多糖(lipopolysaccharide,LPS)诱导的人鼻黏膜上皮细胞(human nasal epithelial cells,HNECs)表达黏蛋白MUC5AC的影响。方法:将体外培养的原代人鼻黏膜上皮细胞分成对照组(C,不予任何处理)、LPS组(LPS,加入LPS 1mg·L-1)、BCQB低剂量组(BCQBL,加入LPS 1mg·L-1及BCQB 10-8 mol·L-1)、BCQB中剂量组(BCQBM,加入LPS 1mg·L-1及BCQB 10-7 mol·L-1)、BCQB高剂量组(BCQBH,加入LPS 1mg·L-1及BCQB 10-6 mol·L-1)和异丙托溴铵组(ipratropium bromide,IB,加入LPS 1mg·L-1及IB 10-6mol·L-1)。经含生长因子的无血清培养基37℃,5%CO2培养24小时后,采用Real-time PCR法检测鼻黏膜上皮细胞中MUC5AC m RNA的表达,Western Blot法检测鼻黏膜上皮细胞中MUC5AC蛋白表达,ELISA法检测上清液中MUC5AC蛋白表达。结果:LPS组与对照组相比MUC5AC m RNA和蛋白表达明显升高(均P<0.01),BCQB各剂量组与LPS组相比MUC5AC m RNA和蛋白表达明显降低(P<0.01,P<0.05),且高剂量组与对照组比较差异无统计学意义(均P>0.05)。结论:BCQB能够抑制LPS诱导的人鼻黏膜上皮细胞黏蛋白MUC5AC表达,提示其可能成为鼻腔黏液高分泌性疾病治疗新药。

【Abstract】 Aim:To investigate the effect of bencycloquidium bromide(BCQB) on mucus MUC5 AC expression induced by lipopolysaccharide in cultured human nasal epithelial cells(HNECs).Methods:Primary culture of human nasal epithelial cells(HNECs) were randomly devided into control group( C,with no treatment), LPS group( LPS,with LPS 1mg·L-1 added in), BCQB low dose group( BCQBL,with LPS 1mg·L-1 and BCQB 10-8 mol·L-1 added in), BCQB middle dose group( BCQBM,with LPS 1mg·L-1 and BCQB 10-7 mol·L-1 added in), BCQB high dose group( BCQBH,with LPS 1mg·L-1 and BCQB 10-6 mol·L-1 added in) and ipratropium bromide group(IB,with LPS 1mg·L-1 and IB 10-6 mol·L-1 added in). After incubation at 37 ℃ with 5% CO2 for 24 h, the expression of MUC5 AC mRNA was detected with Real-time PCR and the expression of MUC5 AC protein in HNECs was detected with Western Blot, while the expression of MUC5 AC protein in supernatant was detected with ELISA in each group.Results:As compared with control group,the expression of MUC5 AC mRNA and protein increased significantly in LPS group(each P <0.01).As compared with LPS group,the expression of MUC5 AC mRNA and protein decreased significantly in each group of BCQB(P <0.01,P <0.05),and there was no statistical difference between BCQB high dose group and control group(each P >0.05).Conclusion:Bencycloquidium bromide can suppress MUC5 AC expression induced by LPS in cultured human nasal epithelial cells,indicating that BCQB may be a new drug for nasal mucous hypersecretion diseases.
