粘附斑激酶在子宫腺肌症中的表达及其与痛经相关性分析

【摘要】 目的:探讨FAK在子宫腺肌症在位内膜及病灶中的表达及其与临床特征的相关性。方法:采用免疫组化、蛋白免疫印迹技术及RT-PCR技术检测22例子宫腺肌症在位内膜及腺肌症病灶组织中FAK蛋白及mRNA的表达,并以29例正常子宫内膜组织作为对照。结果:1.FAK蛋白在子宫内膜腺细胞和基质细胞中均有表达,其表达主要见于细胞浆内。FAK蛋白在子宫腺肌症病灶组织有表达。2.通过灰度分析,子宫腺肌症组FAK蛋白相对表达量为(0.54±0.04),明显高于对照组(0.43±0.05)(P<0.05)。子宫腺肌症组病灶组织中FAK蛋白相对表达量为(0.37±0.04),明显低于对照组(0.43±0.05)(P<0.05)。子宫腺肌症组FAK mRNA相对表达量为(0.46±0.04),明显高于对照组(0.29±0.05)(P<0.01)。子宫腺肌症组病灶组织中FAK mRNA相对表达量为(0.22±0.04),明显低于对照组(0.29±0.05)(P<0.05)。3.合并痛经的子宫腺肌症患者在位内膜中FAK蛋白表达水平也显著高于无此症状者(P<0.05)。通过Person分析发现子宫腺肌症患者病灶组织中FAK蛋白表达水平与痛经VAS评分呈正相关(n=22;r2=0.110,P=0.009)。结论:1.FAK在子宫腺肌症在位内膜组织中表达上调,FAK可能与子宫腺肌症发病有关。2.FAK在子宫腺肌症病灶中表达程度与患者痛经呈正相关,FAK可能与子宫腺肌症的痛经有关。
【Abstract】 Objective:To investigate whether FAK expression is altered in adenomyosis.Methods:FAK protein expression was assessed by immunohistochemistry, western blotting analysis.Results:1. FAK immunoreactive staining was present in the cytoplasm of glandular epithelial and stromal cells in eutopic emdometial tissue and ectopic lesions.2. The anti-FAK antibody detected a band at 125kDa in protein extracts from all endometrial tissues. After normalizing each band of FAK with β-actin from different samples, we found that FAK expression in eutopic endometrial tissues of women with adenomyosis (0.54 ± 0.04) was significantly higher than that of control (0.43 ± 0.05) (P<0.05). FAK expression in ectopic tissues of women with adenomyosis (0.37 ± 0.04) was significantly higher than that of control (0.43 ± 0.05) (P<0.05).3. Semi-quantitative analysis showed that FAK mRNA expression level in eutopic endometrial tissues of women with adenomyosis (0.46 ± 0.04) was significantly higher than that of control (0.29 ± 0.05) (P<0.05). FAK expression in ectopic tissues of women with adenomyosis (0.22 ± 0.04) was significantly higher than that of control (0.29 ± 0.05) (P<0.05).4. Compared with women without dysmenorrhea, FAK protein expression was significantly increased in cases with dysmenorrheal in adenomyosis (P<0.05). A positive correlation was observed between FAK protein expression and dysmenorrhea (r2=0.121,P=0.011;Fig.4) in patients with adenomyosis.Conclusion:1. FAK immunoreactive staining was present in the cytoplasm of glandular epithelial and stromal cells in eutopic endometrium. Expression of FAK was increased in endometrium of women with adenomyosis, and the elevation of FAK expression was associated with dysmenorrhea.2. FAK immunoreactive staining was present in the lesion tissue of adenomyosis. Its expression level was associated with the clinicopathology, which might be related to the progress of adenomyosis.
