天然产物Alsmaphorazine衍生物的全合成

【摘要】 本文的主要是对天然产物Alsmaphorazine E的全合成研究,我们以从自然界中提取的分子Alsmaphorazine E作为目标,运用自身所掌握的有机化学合成相关知识,结合最新的科研理论,运用完善的鉴定手段去完成目标。本论文主要阐述了我们的科研思路以及在研究过程中遇到的主要问题及其解决方案。文章主要分为两个部分:在本文的前一部分,我们介绍了Alsmaphorazine E及其一系列家族成员的背景,其中包含了Alsmaphorazine一系列分子的分离提取及其相对构型和绝对构型的确定,还简单介绍了可能的生物合成途径。目标天然产物的整个结构复杂拥挤,对全合成研究有一定挑战难度。故而,对该分子进行全合成探索具有重要意义。而且该系列分子在自然界中含量较低,分离困难,故而,完成了对Alsmaphorazine E的全合成后,对其及其相关衍生物的生物活性的鉴定,对相关学科皆有重要意义。因此我们对此课题产生了浓厚的兴趣。在本论文的第二部分,我们将着重讲述具体的全合成研究工作,包括逆合成分析、合成进展以及今后的工作计划。首先,对于Alsmaphorazine E我们提出了两种合成策略。两种策略均以吲哚双环骨架作为起始,经过不同的路线,最终都以还原Heck反应关上最后一个含氮六元环,再经一步双键氧化即可得到最终目标产物。策略一主要是通过一个大侧链形成一个大环,再一步关环形成两个小环;策略二则是先在基础骨架上接好两或三个小侧链,依次关上六元环、五元环。最终,两条路线并没有得到彻底的实现,我们以部分成功的步骤作为基础进行修改,欲将目标达成。
【Abstract】 The thesis aims at the studies on the total synthesis of the natural product Alsmaphorazine E which are extracted from nature. We use organic chemical knowledge about synthesis, combining the latest scientific research theory and the use of identification and improved means to accomplish the goal. This paper mainly expounds our research ideas and the main problems encountered in the course of the study and its solution. The article is divided into two parts:In the first part, we introduce the background of Alsmaphorazine E and their family menbers, including how they are extracted and separated and how their absolute configuration are determinated, also introduces some possible biosynthetic pathway to synthesis them.It’s a challenge to synthesis the natural products because their structure are so complex.Therefore, it’s important to explor the molecular total synthesis. And the series of molecularcontent in nature is low, difficult separation, therefore completing the total synthesis of Alsmaphorazine E, identification of its biological activity and related derivatives, have important significance to the related disciplines. Therefore we had a strong interest in this subject.In the second part, we will focus on the work of total synthesis including inverse synthetic analysis, synthesis progress and plans for future work.
