益生菌对雏鸡免疫器官免疫功能及β-防御素-1mRNA表达的影响
Effects of Immune Function And Expression of β-defensin-1mRNA in Immune Organs on Probiotics-Fed Chicks

【摘要】 以1日龄雏鸡为研究对象,应用荧光定量PCR等技术,结合器官组织石蜡切片及组织化学染色等方法,通过免疫器官——胸腺、脾脏、法氏囊鸡β-防御素-1(Gal-1)mRNA表达,浆细胞数量及其核浆比,TANAE+细胞数量等有关指标动态变化的检测,较全面系统的研究了不同种类益生菌对雏鸡免疫器官免疫功能和Gal-1mRNA表达的影响,旨在为益生菌及其制剂在动物生产中的应用及其研发新产品提供科学实验依据,研究结果发现:(1)雏鸡单独应用双歧杆菌后,胸腺、脾脏、法氏囊中TANAE+细胞数量均于7~13日龄时较对照雏鸡明显增多(P<0.05);脾脏、法氏囊和胸腺中的浆细胞数量分别于10~13日龄和7~10日龄时较对照雏鸡明显增多(P<0.05);但Gal-1mRNA表达于10~13日龄时较对照雏鸡明显提高(P<0.05或P<0.01)。(2)雏鸡单独应用乳酸杆菌后,脾脏和法氏囊中浆细胞和TANAE+细胞数量以及Gal-1m RNA均于7~13日龄时显著或极显著高于对照组雏鸡(P<0.05或P<0.01),而胸腺中浆细胞和TANAE+细胞数量于7~10日龄时明显多于对照组雏鸡(P<0.05),但Gal-1mRNA表达于7日龄时较对照雏鸡明显提高(P<0.05或P<0.01)。(3)双歧杆菌和乳酸杆菌联合应用雏鸡后,胸腺、脾脏和法氏囊中的TANAE+细胞数量分别于7~10、13和10~13日龄时明显多于对照组雏鸡(P<0.05);免疫器官中的浆细胞均与7日龄时明显多于对照组雏鸡(P<0.05);但Gal-1mRNA表达于10~13日龄时较对照雏鸡明显提高(P<0.05或P<0.01)。乳酸杆菌对雏鸡免疫器官T淋巴细胞和浆细胞数量及Gal-1m RNA表达的作用效果优于双歧杆菌。
【Abstract】 One day old chicks fed with probiotics were used in this study. The technologies of tissue paraffin section and histochemical staining and real-time fluorescence quantitative PCR were used to respectively research the following details in immune organs—bursa of Fabricius、spleens、thymus of chicks fed with different kinds of probiotics: the plasma cell number and ratio of nucleus, the number of TANAE+ cells and the dynamic changes of Gallinacin-1(Gal-1) m RNA expression. Our objective is to comprehensive understand the influence of different types of probiotics on chicks immune organs immune function and the expression of Gal-1m RNA, and provide scientific experimental basis for the research of probiotics. The results were as follows:(1) Chicks fed with Bifidobacterium, the number of TANAE+ cells in thymus、spleen、bursa were higher than that of the control chicks(P<0.05) at 7~13 days; the number of plasma cells in spleen、bursa and thymus were higher than that of the control chicks(P<0.05) at 10~13days, 7~10days, respectively; while the Gal-1m RNA expression in above immune organs is significantly higher than that of the control chicks(P<0.05 or P<0.01) at 10~13 days.(2) Chicks fed with Lactobacillus, the number of TANAE+ cells、 plasma cells and the Gal-1m RNA expression in spleen and bursa were higher than that of the control chicks(P<0.05 or P<0.01) at 7~13 days, while the number of plasma cells and TANAE+ cells was higher than that of the control chicks(P<0.05) at 7~10 days; while the Gal-1m RNA expression in above immune organs is significantly higher than that of the control chicks(P<0.05 or P<0.01) at 7days.(3) Chicks fed with Bifidobacterium and Lactobacillus in combination, the number of TANAE+ cells in thymus、spleen、bursa were higher than that of the control chicks(P<0.05) at 7~10 days、13days and 10~13days; the number of plasma cell in above immune organs were higher than that of the control chicks(P<0.05) at 7 days.While the Gal-1m RNA expression in above immune organs is significantly higher than that of the control chicks(P<0.05 or P<0.01) at 10~13 days.The effect of Bifidobacterium to the number of plasma cells and TANAE+ cells, and the Gal-1 m RNA expression is better than others.
