食品中合成色素的检测
Determination of Synthetic Pigment in Food

【摘要】 食用色素是一类常用的食品添加剂,在食品行业中食用色素作为赋予和改善食品色泽的食品添加剂而被广泛应用。然而在巨大的经济利益的驱使下,有些商家为了所售商品的美观而超量、超范围使用合成色素,对食品安全造成很大威胁。因此世界各国都对食用合成色素的种类和用量做了严格的规定。本文拟通过优化高效液相色谱法建立食用色素的检测方法,并在此基础上对饮料、膨化食品等样品中色素的含量进行检测,为食用色素的检测奠定基础。主要研究内容如下:高效液相色谱法测定食用色素方法的建立:通过对检测波长、流动相的选择与配比及流速的优化,建立高效液相色谱同时测定胭脂红、苋菜红、日落黄及柠檬黄的方法。以饮料和膨化食品为研究材料,利用高效液相色谱法检测四种合成色素(胭脂红、苋菜红、日落黄和柠檬黄)。采用Agilent XDB-C18柱(4.6 mm×250 mm,5μm)为色谱柱,用甲醇-0.02mol/L乙酸铵溶液进行梯度洗脱,洗脱条件:0~9 min甲醇20%~50%;9~18 min为50%~75%;18~20 min为75%~20%,流速0.80 m L/min,检测波长254 nm。样品中食用色素的测定:以市场上常见的美年达、芬达饮料和膨化食品为样品,按照建立的测定方法分别就样品中含有胭脂红、苋菜红、日落黄和柠檬黄四种色素含量进行测定,从而确定样品中色素的含量是否符合国家标准。该法操作简便,适用于饮料和膨化食品中常见合成色素的快速测定。
【Abstract】 Food pigment is a kind of common food additives. It is widely applied in the food industry. However, some businessmen who had been driven by the huge economic benefits used the excess or ultra range synthetic pigment in order to make the food beautiful. It poses a great threat to food safety. So the kinds and usage of the synthetic pigment are ruled strictly in the world.The detection methods of the food pigment were established by optimization of determination conditions for the high performance liquid chromatography(HPLC). And the pigment was detected in beverage and puffed food. The foundation was laid for the determination of the food pigment. The main researches were as follows.The determination method of food pigment was established by HPLC. The material were four food pigments for carmine, Amaranth, sunset yellow and lemon yellow in beverage and Puffed food. The chromatographic column was Agilent XDB-C18 column(4.6 mm×250 mm, 5 μm).The mobile phase was methanol-0.02 mol/L ammonium acetate.The elution conditions were : flow rates was 0.80 mL/min, the detection wavelength was254 nm, the concentrations of methanol were 20%~50%, 50%~75% and 75%~20% when the times were 0~9 min, 9~18 min and 18~20 min respectively. The method was simple and applicable to rapid determination of the synthetic pigment in beverage and puffed food.The food pigment was determinated in the sample. The samples were Mirinda, Fanta and puffed food. The carmine, Amaranth, sunset yellow and lemon yellow were determinated in the samples by the above method. So the content of pigments in sample may be confirmed whether it was in accord with the national standard.
