三氯蔗糖合成工艺的研究

【摘要】 本文就高甜度甜味剂三氯蔗糖的合成方法及工艺主要进行了以下研究工作:实验以蔗糖为起始原料,分别以苯甲酸甲酯、乙酸乙酯、乙酸乙烯酯、原乙酸三甲酯为酯化剂,在不同催化剂催化下合成蔗糖-6-单酯,通过实验确定蔗糖-6-单酯的最佳合成方法为:以蔗糖为原料,苯甲酸甲酯为酯化试剂,使用碳酸铯为催化剂合成蔗糖-6-苯甲酸酯。探讨了催化剂的用量、反应温度、反应时间和物料配比对反应的影响。最优的反应条件:催化剂的质量分数为2%(以对蔗糖的质量计,下同),n(蔗糖):n(苯甲酸甲酯)=1:4,反应温度为45℃,反应真空度为0.05MPa,反应时间为5h。该条件下,蔗糖-6-苯甲酸酯的收率可达81.0%。蔗糖-6-单酯的选择性氯化也是合成三氯蔗糖的关键步骤,本实验中,用二氯亚砜(SOCl2)与N,N-二甲基甲酰胺(DMF)作用预先生成Vilsmeier试剂,再将蔗糖-6-苯甲酸酯/DMF溶液向其中滴加进行氯化反应。在氯化反应中,通过对影响反应的各因素进行优化,确定了氯化反应的最佳工艺条件为反应温度110℃,SOCl2与蔗糖-6-苯甲酸酯的摩尔比8:1,DMF(mL)与蔗糖-6-苯甲酸酯(g)之比10:1,反应时间2h。用乙酸乙酯作萃取剂,该条件下,三氯蔗糖-6-苯甲酸酯的收率可达60.2%。三氯蔗糖-6-苯甲酸酯在甲醇钠/甲醇溶液中,调节pH为9,反应温度10℃时,92.3%的三氯蔗糖-6-苯甲酸酯可转化为三氯蔗糖。三氯蔗糖的粗产品在水中重结晶两次即得到纯净的三氯蔗糖。经过三步反应,三氯蔗糖总收率为39.2%。
【Abstract】 In this paper, the synthesis method and process of sucralose were used asfollowing work:The sucrose-6-monoesters were synthesized from sucrose and differentesterifying agent such as methyl benzoate, ethyl acetate, vinyl acetate and trimethylorthoacetate with different catalysts. We get the optimal synthesis method throughexperiments: the sucrose-6-benzoate was synthesized from sucrose and methylbenzoate using cesium carbonate as catalyst. It was investigated that the influence ofthe catalyst dosage, reaction temperature, reaction time and material ratio. Theoptimal reactant conditions were: using cesium carbonate as catalyst with a massfraction of2%based on the mass of sucrose, the molar ratio n(sucrose): n(methylbenzoate)=1:4, the reaction temperature45℃, the vacuum degree0.05MPa, and thereaction time5hours. Under the optimal reaction conditions, the yield ofsucrose-6-benzoate was81.0%.Selective chlorination of the sucrose-6-monoesters is also a key step in thesynthesis of sucralose. In this experiment, firstly vilsmeier reagent was produced bythe reaction of SOCl2and DMF, and then the chlorination reaction was carried outthrough sucrose-6-benzoate/DMF was dropped into the vilsmeier reagent. It wasinvestigated that the influence of different factors. The optimal reactant conditionswere: the reaction temperature110℃, the molar ratio n(SOCl2):n(sucrose-6-benzoate)=8:1, the ratio DMF(mL):sucrose-6-benzoate(g)=10:1, and thereaction time5hours. Ethyl acetate is the extraction agent. Under this condition, theyield of sucralose-6-benzoate was60.2%.In the solution of sodium methoxide/methanol,92.3%sucralose-6-benzoate canbe converted to sucralose under the conditions of10°C and pH=9. The crude productof sucralose was recrystallized twice from water to give pure sucralose. Through thesethree steps, the total yield of sucralose was39.2%.
