硫化氢对大鼠深Ⅱ°烧伤创面愈合的影响

【摘要】 目的探究硫化氢在大鼠深Ⅱ°烧伤创面愈合过程的作用及其意义。方法将108只8周龄SD大鼠,雌雄各半,于大鼠背部制成约5%烧伤面积的(深Ⅱo)烧伤模型。然后随机均分为三组,即对照组、硫氢化钠组、PPG组。对照组仅单纯烫伤,每日定时注射1ml生理盐水。NaHS组每日定时腹腔内注射NaHS约1ml(56μmol·kg-1·d-1),PPG组每日定时腹腔内注射PPG45mg·kg-1·d-1。在大鼠烧伤后的第1d、4d、7d、14d、21d和28d,分别详细观察记录他们的创面溶痂比率和创面愈合比率,并在各时相去病理标本做病理切片后行组织病理学平分。结果(1)烧伤后第7、14天,NaHS组创面溶痂率明显低于PPG组及对照组,其余各观察时相无明显差异,(2)烧伤后第14、21天,NaHS组创面愈合比率高于对照组和PPG组,而PPG组创面愈比率合低于对照组和NaHS组,其余各时相无统计学差异(3)NaHS组创面组织病理学评分于烧伤后第14及21天高于对照组,于伤后第7、14及21天高于PPG组;PPG组于14及21天组织病理学评分均低于对照组。结论硫化氢参与了大鼠深Ⅱ°烧伤创面的愈合过程,应用外源性H2S后,创面溶痂时间推迟,创面愈合时间缩短,提示硫化氢可能促进烧伤后皮肤的痂下愈合。

【Abstract】 Objective To study the function and the significance of sulfuretted hydrogenin the process of healing of burn wound. Methods108healthy and eight weeks of rats(female and male were half-and-half) which were made a burn model in the back,wererandomly divided into3groups:control group,sodium hydrosulfide group(NaHS) andLD-propargylglycine group(PPG). The control group were injected0.9%sodiumchloride1ml once a day.NaHS group were injected NaHS56μmol·kg-1·d-1and the PPG groupwere inject-ed PPG45mg·kg-1·d-1.Eschar dissolution ratio,wound healing rate andhistopathological score would be recorded after1d,7d,14d,21d and28d fromburn.Results (1)NaHS group’s eschardi-ssolution ratio were lower than control groupand PPG group on the7th and14thd-ay after the rats were burned. The difference resultof other time phase was not statistically significant.(2)NaHS group’s wound healingrate was higher than con-trol group’s and PPG group’s,while PPG group’s wound healingrate was lower than NaHS group’s and control group’s.The difference result of othertime phase was n-ot statistically significant.(3)NaHS group’shistopathological scoreis higher th-an control group’s on the14th and21th days after that the rats wereburned. It’also higher than PPG group’s on the7th、14th and21th days.PPG group’shistopa-thological score is lower than control group’s on the14th and21th days aftert-he rats were burned.Conclusion The H2S/CSE system has participated in the healingprocess of deep partial thickness burn wound in rats. Extrinsic H2S can shorten thewound healing time and delay eschar dissolution time.It’s showed that raising thesulfuretted hydrogen can promote burn wound healing under the eschar.
