消化法检验旋毛虫最适条件的筛选

【摘要】 旋毛虫病是一种人兽共患寄生虫病,呈世界性分布,目前在一些发展中国家(如泰国、阿根廷及中国等)仍常有暴发,其不但危害人类公共卫生,而且成为影响猪肉生产和食品安全的重大经济问题之一。由于屠宰动物的旋毛虫病检验是目前控制该病向人类传播的唯一有效策略,因此该病的检验已成为世界各国更是世界动物卫生组织(OIE)法规规定的屠宰动物强制性、首检和必检病种(我国始于1956年)。目前已确定旋毛虫有8个种和4个基因型,其检测方法一律采用集样消化法。由于旋毛虫国际消化法采用统一标准和规定,但实际上旋毛虫不同种与基因型存在不同的生物学特征,因此在消化和收集方法上应存在一定的差异。本研究以小鼠为动物模型,利用实验室保存传代的旋毛虫国际标准的8个种和4个基因型感染Balb/c小鼠,感染40天后检测其繁殖能力指数(RCI)和每克肌肉幼虫数(LPG);进一步分别在4℃和25℃条件下计算不同时间点的回收肌幼虫百分比,比较和分析旋毛虫不同种及基因型对小鼠繁殖能力指数、沉降速率及消化法检验最适条件的差异。结果表明:用消化液原液计数,得出的RCI值更为准确,有望改写现有教科书及相关研究报道中有关旋毛虫繁殖力指数的经典记载;计算不同温度不同时间点收集到的肌幼虫数目与消化液原液总的肌幼虫数目的比值,并计算出回收肌幼虫达到100%时的沉降速率,比较发现旋毛虫不同种及基因型在不同温度和沉降时间上,回收的肌幼虫百分比存在明显差异,确定出适合已知旋毛虫每个种和基因型的消化法检验最适条件。因此,本研究结果为旋毛虫国际消化法的改善提供了重要的参考数据。

【Abstract】 Nematodes of the genus Trichinella are zoonotic parasites with a cosmopolitandistribution. Worldwide, Trichinella spp. have been found to be the causative agentsof human trichinellosis, a disease that not only is a public health hazard by affectinghuman patients but also represents an ecomomic problem in porcine amimalproduction and food safety. The test trichinellosis of slaughter animals is currently theonly effective strategies to control the disease transmission to human, so theinspection of the disease has become first check, which has been rules and regulationsby the World Organization for Animal Health/Office International Des Epizooties(OIE), our country started in1956. Trichinella international digestion method makeuniform standards and rules, however, according to different biological characteristicsof different species from Trichinella spp, therefore the digestion and collectionmethods should exist a certain differences.On this study: mice were used as animal model, larvae recovered after40dayspost infected were used to count RCI (Reproductive capacity index) and LPG (Larvaeper gram muscle tissue). In this experiment, mice were infected with8Trichinellaspecies and4genotypes, which have been saved and extended in our lab, the RCI andLPG were counted, and then analysised by statistics; the subsidence coefficient underthe condition of4℃and25℃at different time points were calculated, and thencompared and analysised the infect abilities and Sedimentation coefficient of differentTrichinella species and genotypes in mice. The results show the digestiveconcentrate to count RCI and LPG is precicer than reported ones, it is likely to rewriteexisting records; and at different time and temperature points, the sedimentationcoefficients are different depend on different Trichinella species and genotypes, wevalue the optimized digestion method for Trichinella spp, which should provide areliable experimental reference data for the improving Trichinella internationaldigestion method.
