川西獐牙菜化学成分研究

【摘要】 川西獐牙菜(Swertia mussotii Franch.)是龙胆科(Gentianaceae)獐牙菜属(swertia)植物,是“藏茵陈”类药用植物的基原植物。川西獐牙菜为1年生草本植物,主要分布在西藏、云南、四川西北部、青海西南部等海拔1900m-3800m的地区。具有清热解毒,利胆退黄之功效,全草用于治疗肝胆系统疾病。川西獐牙菜中含有的酮类化合物是最主要的药用成分基础,另含有獐牙菜苷(sweroside)、龙胆苦苷(Gentiopicroside)等环烯醚萜类化合物,该化合物常用于保肝类药物中,也是龙胆科植物的代表性成分。本实验通过正交实验法优选川西獐牙菜提取工艺,采用L16(45)正交实验设计考察单因素对川西獐牙菜有效成分提取率的影响。实验设定乙醇浓度、提取次数、料液比、提取时间、提取温度5个因素4水平对川西獐牙菜的提取工艺进行研究。并通过HPLC法测定其獐牙菜苷及龙胆苦苷的含量以优选提取工艺。结果表明收率与龙胆苦苷含量上各工艺无显著差异,而用90%的乙醇作为提取溶剂,料液比为1:24,微沸,回流提取3次,每次4h为有效成分獐牙菜苷提取的最佳工艺,经验证该工艺条件稳定可行。本实验采用硅胶和聚酰胺柱色谱进行分离纯化,根据化合物的理化性质和波谱数据鉴定其化学结构,从川西獐牙菜的脂溶性部分分离得到4个化合物,经鉴定为1-羟基-3,7,8-三甲氧基(?)酮;1-羟基-2,3,4,5-四甲氧基(?)酮;1,8-二羟基-3,7-二甲氧基(?)酮;1,7,8-三羟基-3-甲氧基(?)酮。
【Abstract】 Swertia mussotii Franch. is Gentianaceae swertia plant and one of Tibetan medicine Zangyinchen plants.as one-year plant, distributed at an altitude of1900-3800m in Tibet, Yunnan, west northwest of Sichuan, and Southwest of Qinghai, is the raw material of medicinal used in the treatment of diseases of the liver and gallbladder system. The compounds containing in Swertia mussotii include ketone, sweroside, gentiopicroside etc.The L16(45) orthogonal experiment design and single factor analysis were utilized to select the most effective technology of Swertia mussotii compound extraction. Four levels and five factors including ethanol concentration, extraction times, material liquid ratio, extraction time, and extraction temperature were designed and studied. HPLC method was used to analyze content of the sweroside and gentiopicroside. Results showed that90%ethanol as extraction solvent, material liquid ratio1:24, boiling, reflux extracting3times, and4h were one of the most effective technology, which was stable and feasible.This experiment was studied with the polyamide and silica gel chromatography for separation and purification. According to the physical and chemical properties and spectral data, four kinds of compounds which were1-hydroxy-3,7,8trimethoxyxanthone,1-hydroxy-2,3,4,5-tetramethoxyxanthone,1,8-dihydroxy-3,7dimethoxyxanthone,1,7,8-trihydroxy-3-methoxyxanthone were separated and identified.
