灵芝首乌酒的研制及其体外抗氧化活性的研究

【摘要】 本实验旨在以灵芝、制何首乌等为主要原料,当归、熟地等为辅助原料,研制出一种色香味较佳的灵芝首乌酒。通过对各中药材的浸泡工艺、有效成分提取工艺、勾兑调味方案进行优化,获得了质量稳定,风味较优的产品。同时对成品酒进行了初步的抗氧化活性研究及其缓解痛经的效果的探讨。主要结果如下:1.以灵芝的主要活性成分灵芝多糖的提取率来确定灵芝的浸泡方法,比较灵芝水提和醇提浸膏中灵芝多糖含量得出,水提浸膏中灵芝多糖含量远高于醇提浸膏。因此制备灵芝浸酒液可以先用乙醇浸提,醇溶性物质浸出后,滤渣进行水提,制成水浸膏溶于低度酒中,可以充分利用灵芝中有效成分。2.何首乌中的有效成分二苯乙烯苷的浸提优化得出:液固比为20:1,酒精体积浓度为50%,浸提时间为5天,分两次浸提,可以得到最大浸提率,在此条件下,何首乌浸出液中可溶性固形物含量也达到较佳水平。以何首乌浸提条件为基础,研究了其他几种中药材浸出液中可溶性固形物含量与时间的关系,得出它们的浸提时间为4~5天,可溶性固形物含量可达到较佳水平。3.通过正交实验对各浸泡液勾兑混合,得出:灵芝浸酒液、何首乌枸杞混合液和其他中药材的混合体积比为3:5:5,可得到色泽、透明度及风味较优的混合酒液。澄清后的混合酒液进行正交实验调味得出,蔗糖添加量为8%,柠檬酸添加量为0.4%,酒精度(v/v)45%。并对成品酒进行了理化及卫生指标的检测。4.测定灵芝首乌酒清除DPPH自由基、羟自由基、ABTS自由基、超氧阴离子自由基的能力及其还原能力,以抗坏血酸为对照,结果得出:灵芝首乌酒对以上自由基均有清除效果,并且具有较强的还原能力。5.通过对20例痛经患者服用灵芝首乌酒的疗效观察得出,灵芝首乌酒对痛经具有一定的缓解作用。

【Abstract】 This thesis was aim at using Ganoderma Lucidum and Polygonum multiflorum Thunb asmain materials, complemented with Radix Angelicae Sinensis, Rehmannia Glutinosa Libosch.etc. to produce liquor with good appearance, taste and aroma, meanwhile putting forward thefeasible manufacture technology. The soaking process of Chinese herbal medicine, the extractof effective components, the scheme of blending were optimized. Finally, a kind of liquorwith stable quality and acceptable flavor was attained. The antioxidant activities of the liquorwere evaluated.The effect of alleviating dysmenorrheal were discussed. The following resultswere obtained.Water extraction and alcohol extraction were applied to the Ganoderma Lucidumpolysaccharides from Ganoderma Lucidum. The results showed that the higher content ofGanoderma Lucidum polysaccharides in water extraction. In order to get higher qualityGanoderma Lucidum immersion of spirit, Ganoderma Lucidum was immersed in alcohol toget alcohol leaching extract first, and then the residue was immersed in water, concentrated toextractum, dissolving water extract soluble in low-alcohol wine. Through this way,Ganoderma Lucidum could be used fully.The optimal conditions to extract the effective ingredient stilbene glycoside inPolygonum multiflorum Thunb were as follows: temputerature of30℃,50%(V/V) of alcohol,20:1solid/liquid ratio and soaking twice for5days. Under this condition, the soluble solidscontent of Polygonum multiflorum Thunb leaching also were reached at a better level.The different kinds of Chinese herbal medicines were extracted based on the condition ofPolygonum multiflorum Thunb, obtaining the soak of the medicinal material leaching time for4~5days, in this case, the soluble solids content could reach at better level.Different leach liquors were optimized by an L9(34) orthogonal test. Results showed thatthe extract of Ganoderma Lucidum:(Polygonum multiflorum Thunb and Lycium barbarum):other Chinese herbal medicines=3:5:5(v/v/v), and available color, transparency and flavormixtures were attained. The clarified mixtures were optimized by orthogonal test. The results showed that theoptimal blending Ganoderma Lucidum and Polygonum multiflorum Thunb wine spirits wereas following:45%(V/V) of alcohol, citric acid0.4%,cane sugar8%.Then, the variousphysical and chemical index of the wine were detected.The antioxidant properties of wine were evaluated by using different antioxidation tests,including reducing power, DPPH radical scavenging, ABTS free radical, superoxide anionradical scavenging, hydroxyl radical scavenging. The results showed that the wine exhibitedstrong reducing capacity, DPPH radical scavenging, ABTS free radical, superoxide anionradical scavenging, hydroxyl radical scavenging. The wine has good antioxidant ability.Meanwhile, the wine has good effect on dysmenorrhea in remission.
