高粱醋皂甙提取工艺及其对家兔血小板聚集的影响

【摘要】 食醋是中国传统的发酵酸性调味食品,深受中国人民的喜爱,其拥有的抗血栓、调节血糖和脂质代谢、缓解疲劳、抗氧化、促进食欲等多种对人体有益的生物活性功能也受到了人们的广泛重视。高粱醋在发酵过程中会产生多种对人体有益的生物活性物质,而皂甙就是其中一种具有很强抗血栓生理活性的物质。为探寻高粱醋中皂甙的提取工艺以及对家兔血小板聚集的影响,本研究选用了超滤膜分子筛技术、有机溶剂萃取技术、大孔树脂分离技术、葡聚糖凝胶分离技术,将高粱醋提取、分离、纯化得到皂甙提取物。实验结果表明,在有机溶剂萃取过程中使用75%乙醇作为浸提液、超声提取30min、提取温度为50℃时,在大孔树脂静态吸附过程中收集75%乙醇部分,在葡聚糖凝胶层析过程中使用去离子水作为流动相,得到纯度较高的皂甙提取物。并使用ADP与胶原作为诱导剂,以兔血为血源,实验证明高粱醋中皂甙提取物具有很强的抗血小板聚集功能,且以ADP为诱导剂的IC50值为3.86mg/ml、以胶原为诱导剂的IC50值为4.57mg/ml。并在皂甙提取物中定量检测总皂甙含量,通过SPSS数据分析得到:皂甙提取物中总皂甙的含量与其抑制抗血小板聚集活性有显著相关性(P<0.01)。可知高粱醋中皂甙提取物对抑制家兔血小板聚集有较强活性。
【Abstract】 As a traditonal fermented acidic seasoning food, Chinese vinegar is a variety of bioactivity, including antithrombotic activity, regulation of blood glucose and lipid metabolism, fatigue relieving, antioxidant activity, stimulation of the appetite. The fermentation process will endow Sorghum vinegar a variety of biologically active substances, such as saponin.This study focused on the extraction and antiplatelet activity of saponins from aged sorghum vinegar. To extract the saponin from the aged vinegar, we compared extraction effects of different isolation materials, including ultrafiltration membrane molecular sieve, organic solvent extraction, macroporous resin, and sephadex. High purity saponin was finally obtained by using organic solvent extraction(75%ethanol plus ultrasonic extraction for30min, at50℃), followed by collecting the elute of75%ethanol in macroporous resin static adsorption process, and Sephdex chromatography(deionized water as the mobile phase). The saponin extract had a strong anti-rabbit platelet aggregation induced by ADP or collagen. The IC50value of saponin to inhibit the platelet aggregation was3.86mg/ml for ADP and4.57mg/ml for collagen. The saponin in the extract was found to be significantly correlated to the anti-platelet aggregation activity(P<0.01). This study showed that saponins in the aged soghum vinegar were of healthy benefit.
