周围神经慢性卡压损伤过程中骨骼肌内SMADS蛋白表达规律的实验研究

【摘要】 目的研究大鼠坐骨神经慢性卡压损伤后,该神经所支配的骨骼肌中Smad的表达规律。方法随机选取56只SD大鼠,采用经典的Mackinnon坐骨神经慢性卡压损伤模型,随机分成正常组和卡压组。分别在正常组、慢性卡压损伤模型术后第0、2、4、8、12、16、20、24周,取大鼠坐骨神经纤维支配的骨骼肌,采用HE染色、RT-PCR和免疫组化等实验技术,检测正常组和慢性卡压损伤组在不同时间段大鼠骨骼肌形态改变、Smad2、Smad3和Smad7阳性细胞的分布规律及其规律。结果1.正常大鼠的腓肠肌中纤维的中有少量Smad阳性表达,阳性颗粒均匀分布于胞浆中,细胞膜周围阳性表达较少,组织间隙明显表达。2.坐骨神经慢性卡压损伤过程中,卡压组腓肠肌中Smad2、Smad3含量呈现先增加后缓慢减少并趋于稳定的状态,但最终略高于正常值;Smad7在腓肠肌中的表达呈现逐渐下降的趋势,最终低于正常值。结论周围神经发生慢性卡压性损伤过程中,Smad2、Smad3和Smad7可能通过TGF-β/Smad信号转导通路的表达,调控其支配骨骼肌的纤维化过程。

【Abstract】 Objective To investigate the expression of SMADS in skeletal muscle during the normal rats with chronically compressed sciatic nerve injury.Methods56SD rats were randomly divided into two groups, normal group and the compressed group. Rats in compressed group were subjected to classical Mackinnon chronic sciatic nerve compression injury. After0、2、4、8、12、16、20、24weeks post-operative, the skeletal muscle dominated by rats’s sciatic nerve from these two groups were dissected. By use of HE staining, RT-PCR, Immunohistochemistry, and other experimental methods, we analyzed the SMADS levels and immunostaining positive neurons in normal rats and the chronic compression injury models.Results1. There was few SMADS positive expression in the gastrocnemius of normal rats.the muscle strength obviously reduced, and the positive particles were evenly distributed between cytoplasm and nucleus. Less positive express were found around cell membranes, and no obvious expression in tissue space.2. The level of Smad2, Smads in the compressed lateral gastrocnemius firstly increased, then slowly reduced and trend to a steady state. But finally it’s over the normal level. Whereas the expression of Smad7in the gastrocnemius trend to a gradual decline.Conclusion There was a few expression of smads in the normal skeletal muscle.When the peripheral nerves were chronically compressed injury, Smad2, Smad3and Smad7regulate and control the skeletal muscle fibrosis process,through the expression of TGF-β/Smad signal transduction pathway.
