NNMT在乳腺癌中的表达及其与临床病理因素关系的初步研究

【摘要】 目的研究NNMT在乳腺癌中的表达水平,分析NNMT与乳腺癌临床病理因素间的关系,探讨NNMT在乳腺癌治疗及预后上的意义。方法1.选取2007年6月-10月于本院乳腺外科就诊且术前未经治疗的乳腺癌患者42例,获得患者临床资料包括:年龄,肿瘤大小,分化,组织分类,淋巴结转移情况,免疫组化(ER、PR和CerbB-2、P53)结果,术前血清肿瘤标志物(CEA、 CA153)结果。2.将42例乳腺癌患者作为研究组,调出术后保存的病理蜡块切玻璃白片做NNMT的免疫组化,抗体为本课题组自行制备的抗NNMT1E-7抗体。选取其中20例标本的癌旁组织作为对照组。3.检测42例乳腺癌和20例癌旁组织的NNMT免疫组化结果(NNMT的阳性率),并与临床病理参数进行统计学分析,研究NNMT与乳腺癌临床病理因素间的关系,探讨NNMT在乳腺癌研究中的意义。结果1.NNMT在乳腺癌组织中有较高的表达水平,阳性率达73.8%;在癌旁组织中表达较低,阳性率为25%,两者间存在显著性差异。2.在阳性的乳腺癌组织中NNMT表达呈弥漫状分布,定位于细胞胞浆和胞核,主要表达在细胞胞浆中。3. NNMT表达水平在乳腺癌患者年龄,肿瘤大小,分化,组织分类,淋巴结转移,ER, CerbB-2的分组比较中未达到统计学差异。但与PR以及P53相关。结论NNMT在乳腺癌中存在过表达并与PR以及P53表达相关,它在乳腺癌治疗和预后上的意义将值得期待,同时可尝试作为一种新的乳腺癌血清学诊断标志物。

【Abstract】 ObjectiveTo study the expression of NNMT and its relationship with clinicopathologic features in breast cancer.MethodsWe studied42patients with breast cancer and collected their clinical data including: age, tumor size, histology, edmondson grade, lymph node metastases, expression of ER, PR, CerbB-2, P53and serum levels of CEA and CA153.The42patients’ postoperative pathologic wax blocks were selected as study group and20cancer side tissues from the42patients were selected as control group. The expression levels of NNMT in both groups were detected by immunohistochemistry. The relationship between the expression of NNMT and the clinicopathologic features of breast cancer was analyzed to investigate the possible role on NNMT in patients with breast cancer Results1. The expression of NNMT protein in breast cancer tissues (73.8%) was significantly higher than that in the cancer side tissues.2. NNMT protein was found to be diffusely expressed both in cytoplasm and nucleus of breast cancer tissues, but mainly in the former.3. No significant association were found between NNMT expression and age, size of tumor, histology, edmondson grade, lymph node metastases, levels of ER, CerbB-2of breast cancer patients(P>0.05). Interestingly, we observed that NNMT expression was positively correlated with PR and P53status.ConclusionsNNMT is over-expressed and associated with PR and P53status in breast cancer, which may become as a possible tumor marker indicating diagnosis and treatment of breast cancer.
