阿苯达唑及其脂质体治疗绵羊肝包虫病效果比较

【摘要】 为进一步验证并比较阿苯达唑脂质体(L-ABZ)与阿苯达唑(ABZ)两种药物治疗绵羊肝包虫病的疗效,本文设计了阿苯达唑脂质体和阿苯达唑两种药物治疗绵羊肝包虫病效果的比较试验,于试验绵羊人工感染包虫后,根据不同药物分为两组,并在不同时间点对各组绵羊的外周血液白细胞(嗜酸性、嗜中性、嗜碱性粒细胞、淋巴细胞、单核细胞)分类计数;分别用流式细胞仪和全自动生化分析仪(FCM)对CD4+T、CD8+T、B淋巴细胞、谷丙转氨酶和血清总蛋白水平进行检测,分析不同药物治疗肝包虫病前后不同时间段绵羊细胞免疫等指标的变化规律,比较阿苯达唑及其脂质体治疗绵羊肝包虫病的效果,同时,制作并观察病理组织切片,进一步探讨两种药物的治疗效果,为治疗绵羊肝包虫病临床用药提供依据。试验一:将从犬小肠中收集分离得到的细粒棘球绦虫(带孕节)给试验绵羊口服人工感染,每只绵羊口服10个成虫孕节。感染当天为试验0d,其后于45d、60d采集绵羊全血分离血清,用ELISA试剂盒检测细粒棘球蚴抗原并判断绵羊感染肝包虫病情况;选择肝包虫病阳性羊6只,分为两组,每组3只,分别口服阿苯达唑和阿苯达唑脂质体,连续用药30天,在-15天、0天、15天、30天分别采集血样,利用流式细胞术等对血液中CD4+T淋巴细胞、CD8+T淋巴细胞、B淋巴细胞、白细胞总数、淋巴细胞、中性粒细胞以及谷丙转氨酶和血清总蛋白水平进行检测。结果显示:给药15天, CD8+T淋巴细胞百分率L-ABZ组显著低于ABZ组(P<0.05);L-ABZ组嗜中性粒细胞水平显著高于ABZ组(P<0.05);在给药30天,ABZ组血清总蛋白水平显著高于L-ABZ组(P<0.05)。给药前后,ABZ与L-ABZ组之间CD4+T淋巴细胞、B淋巴细胞、白细胞总数、淋巴细胞、谷丙转氨酶没有显著的差异(P>0.05)。表明,L-ABZ作用优于ABZ,对绵羊肝包虫病治疗具有积极作用。试验二:选用同期人工感染肝包虫病羊,共分为3组,每组3只,对照组、口服阿苯达唑组(ABZ)、口服阿苯达唑脂质体组(L-ABZ),连续用药30天后屠宰并观察肝脏包虫囊外观及病理组织学变化,并进行疗效的综合评价。结果表明:口服阿苯达唑脂质体治疗绵羊肝包虫病的效果优于单纯口服阿苯达唑。

【Abstract】 To authenticate and compare the therapeutic effect of liposomal Albendazole and Albendazole on Echinococcosis sheep The paper was designed to compare the therapeutic efficacy of liposomal Albendazole and Albendazole on Echinococcosis sheep. After artificial challenge ,the tentative sheep were divided into two groups by different drug, and differential count on sheep peripheral blood leucocyte(acidophilia、neutrophilia、basophilia、granular cell、lymph cell、histoleucocyte)of each group at different time points; to detect CD4+T、CD8+T、B lymph ce ll、glutamate-pyruvate transaminase and total serum protein level by Flow Cytometry(FCM), and analysis the change rule about the index of humoral immunity、mediated immunity of sheep when to cure the echinococcosis by different drug at different time , compare the therapeutic efficacy of liposomal Albendazole and Albendazole on Echinococcosis sheep, at the same time , make manufacture and observe histopathologic slide, contrast the therapeutic efficacy of two drug and provide base for clinical medication.In the first experiment, we infected 10 sheep artificially with the eggs of Echinococcus granulosuse ,which were obtained from the experinment dogs. The day of inoculation is 0d, then abruptio blood serum from sheep whole blood on 45d and 60d,we examined the infection of E. granulosus sheep quickly and exactly by the ELISA diagnose Kit . we infected 6 sheep artificially. They were divided into 2 groups with 3sheep of each one: one group was orally given Albendazole and the other was given liposomal Albendazole for 15d and 30d,then collect blood sample. This study evaluates the therapeutic effect through the blood Immunization indicators such as:CD4+T lymphocytes, CD8+T lymphocytes, B lymphocytes, total leukocytes, lymphocytes, neutrophils, as well as alanine aminotransferase and serum total protein level by Flow cytometry. Results showed that the administration of 15 days later,CD8+T lymphocyte level of ABZ group was significantly higher than that of L-ABZ group , (P<0.05); the neutrophils of L-ABZ Group was significantly higher than that of ABZ group (P<0.05); 30 days later, the serum total protein level of ABZ were significantly higher than L-ABZ group (P<0.05). Before and after administration, there was no significant difference in CD4+T lymphocytes, B lymphocytes, total leukocytes, lymphocytes and alanine aminotransferase between ABZ and L-ABZ (P>0.05),which indicated that the effect of L-ABZ was superior to that of ABZ ,playing an energetic role in curing Ovis aries echinococcosis and calcifying Ves.The second experiment: the homeochronous artificial infection sheep were divided into 2 groups with 3 sheep for each one:Po-ABZ and Po-L-ABZ group by mouth for 30d . After the administration of 30 days Continuously,we observed the the change of the hydatid’s shape and pathologic histology and evaluated the therapeutic effect. The result indicated that therapeutic effect in liposomal Albendazole group is better than Albendazole group.
