葡萄籽原花青素对大鼠肾缺血再灌注保护作用研究

【摘要】 目的:探讨葡萄籽原花青素(GSPE)对大鼠肾缺血-再灌注损伤的保护作用及其机制。方法:选用健康雄性Wistar大鼠36只,随机分为3组:对照组(Sham组)、缺血-再灌注损伤模型组(RIRI组)、葡萄籽原花青素预处理组(GSPE组),建立大鼠肾缺血-再灌注损伤模型,于肾缺血-再灌注2h、2411分别检测血清肌酐(SCr)、血清尿素氮(BUN)、血清一氧化氮(NO)含量的变化和诱生型一氧化氮合酶(iNOS)活性的变化及血清和组织丙二醛(MDA)含量及谷胱甘肽过氧化物酶(GSH-Px)活性的变化,检测肾组织中细胞黏附分子-1(ICAM-1)的表达水平,并观察肾组织病理改变。结果:与对照组相比,模型组和用药组血清SCr、BUN、NO含量升高(p<0.01),iNOS活性升高(p<0.05),血清及肾组织MDA含量升高(p<0.01),GSH-Px活性降低(p<0.01);与模型组相比,用药组的SCr、BUN、NO含量降低(p<0.01),iNOS活性降低(p<0.05),血清及肾组织MDA水平降低(p<0.05), GSH-Px活性升高(p<0.01);免疫组织化学显示,模型组中ICAM-1表达较对照组明显增多,用药组表达则较模型组明显减少;肾脏病理显示用药组肾组织损伤病变较同期模型组明显减轻。结论:在肾缺血-再灌注损伤过程中,葡萄籽原花青素能改善肾肾功能,其机制可能与抗自由基损伤、减轻脂质过氧化及抑制炎症级联反应有关。
【Abstract】 Objective:To investigate the Anti-oxidation Effects of grape-seed proanthocyanidins extrac(GSPE) on Renal Ischemia Reperfusion Injury in Rats.Methods:Thirty-six Wistar rats weighting(180-220g)were divided into three groups randomly:sham operation group(Sham group),renal ischemia reperfusiona injury group(RIRI group), grape seed proanthocyanidins group(GSPE group).There were 6 rats in each group.The content variation of serum creatinine(Scr)、blood urea nitrogen(BUN)、nitric oxide(NO) in plasma and the activity variation of inducible nitric oxide synthase(iNOS) in plasma,the content variation of MDA in plasma and in tissue and the activity variation of GSH-Px in plasma and in tissue,the intercellular adhesion molecules-1(ICAM-1)expression were detected respectively in renal ischemia-reperfusion 2h,24h as well as pathological changes in the different time were also noted by light microscope. Results:Compared with the Sham group,the contents of Scr、BUN、NO and iNOS in plasma and MDA in plasma and in tissue significantly were higer in RIRI group and in GSPE group(p<0.05),and activity of GSH-Px was lower in RIRI group and in GSPE group(p<0.05); Compared with the RIRI group, the contents of Scr、BUN、NO and iNOS were lower in GSPE group,the contents of MDA in plasma and in tissue were also lower (p<0.05);and the activity of GSH-Px were step up (p<0.01);expression of ICAM-1 in rat kidneys in the GSPE group was significantly decreased than that of the RIRI group(p<0.05).The renal pathological changes were more severe in RIRI group than that of GSPE group,and GSPE2h group had no significant difference compared with GSPE24h.Conclusion:The GSPE played a protective role on the kidney in the process of renal ischemia reperfusion injury,the mechanism may be related to anti-free radical injury and reducing the lipid peroxidation.
