金柑总黄酮生理活性研究

【摘要】 金柑属于芸香科柑桔属,富含黄酮,挥发性油,柠檬苦素,膳食纤维以及多种维生素。具有降低胆固醇,降低血压,提高免疫力等多种生理作用。但是,到目前为止,还没有人对这方面进行过研究,这在一定程度上阻碍了金柑产业的开发。本文对金柑总黄酮的生理活性作用进行了研究,本研究的完成将为金柑的开发提供理论上的支持,并会在一定程度上促进金柑产业的开发。本研究以金柑总黄酮为原料,以白鼠为实验对象,探讨不同产地金柑(湖南浏阳和江西遂川,品种为金弹Fortunella Crassifolia Swingle)对其生理活性的影响。按体重配制20mg/kg·bw,80 mg/kg·bw,160 mg/kg·bw三个剂量,同时设空白对照组和模型对照组,主要研究内容及结果如下:以血清SOD和MDA含量、生存寿命、平均寿命、最长寿命为评价指标,研究金柑总黄酮的抗氧化及抗衰老作用。结果表明:与模型对照组相比,实验组能够明显延长小鼠的生存寿命。两中剂量组对血清SOD, MDA含量,平均寿命的影响具有显著性(P<0.05),两高剂量组其影响具有极显著性(P<0.01)。以血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、动脉硬化指数为指标,来评价金柑总黄酮调节血脂作用。结果表明:和模型对照组相比,浏阳金柑中、高剂量组对其影响(除了TG)具有显著性(P<0.05),遂川金柑中、高剂量组对它们的影响(除了TG)具有极显著性(P<0.01)。两中剂量组对TG影响具有显著性(P<0.05),两高剂量组对TG影响具有极显著性(P<0.01)。以对糖尿病小鼠血糖、糖耐量、以及正常小鼠血糖影响为评价指标,来研究金柑总黄酮调节血糖作用。结果表明:和模型对照组相比,灌胃6d后,两个高剂量组对糖尿病小鼠血糖值影响具有显著性(P<0.05),灌胃金柑总黄酮17 d后,两中剂量就可起到改善糖耐量作用,两高剂量组对糖尿病小鼠血糖值影响具有极显著的差异性(P<0.01),各剂量组对正常小鼠的血糖值影响不具有显著差异性。以对巨噬细胞吞噬功能、二硝基氟苯(DNFB)诱导小鼠耳肿胀、游泳时间、耐缺氧能力、胸腺和脾脏指数影响为评价指标,来评价其对免疫系统影响。结果表明:和模型对照组相比,两个中剂量组对其影响具有显著性(P<0.05),两个高剂量组对其影响具有极显著性(P<0.01)。以金柑总黄酮对大肠杆菌,金黄色葡萄球菌抑菌作用以及最大耐受量为评价指标,来评价金柑总黄酮体内抑菌作用以及其安全性。结果表明:和模型对照组相比,金柑总黄酮对大肠杆菌,金黄色葡萄球菌具有明显抑制作用,其最大耐受量为人体摄入量的600倍。以对胃排空、小肠推进率、小肠吸收功能为评价指标,来评价金柑总黄酮对小鼠胃肠吸收作用影响。结果表明:和模型对照组相比,两个中剂量组对其影响具有显著性(P<0.05),两个高剂量组对其影响具有极显著性(P<0.01)。这说明金柑总黄酮具有改善小肠胃肠吸收功能的作用。通过以上研究表明,金柑总黄酮具有抗氧化,调节血脂,调节血糖,提高免疫力,抗菌,促进胃肠吸收的作用,是一种很安全的功能成分,并且生长环境对其生理作用具有很大的影响。
【Abstract】 Kumquat (Fortunella Swingle) is rich in flavone, volatility oil, limonoids, edibility fibre and many kinds of vitamin. It has many kinds of physiology effect such as reduce cholesterin, lower blood pressre, improve immunity. But, up to now, there were no people study on it, this encumbranced the kumquat industrial exploiture at a certain extent. This paper would study on the physiology effect of kumquat total flavone, the accomplishment of this study would provide the theoretic aegis for exploiture kumquat and accelerate kumquat industrial exploiture.In this paper, kumquat total flavone was used as material, mice as experiment object, discussing the effect of different locality kumquat (Hunan Liuyang and Jiangxi Suichuan, the breed is Fortunella Crassifolia Swingle) to the physiology effect of kumquat total flavone. According to body weight, the paper put up three doses:20 mg/kg-bw,80 mg/kg-bw,160 mg/kg-bw, mealwhile, set up blank group and model group. The main study contents and results as follows:The anti-oxidative and anti-decrepitude effect of kumquat total flavone was studied, through evaluating the content of SOD and MDA in serum, living life-span, average life-span and Max. life-span. The results shown:compared with model group, the experiment groups could obvious prolong the life-span of mice. The two middle doses had markedness (P<0.05) effect to SOD, MDA and average life-span, the two higher doses had greatly markedness(P<0.01) effect.The moderating hyperlipaemia of kumquat total flavone was studied, through evaluating content of TC, TG and HDL-C in serum, the index of arteriosclerosis. The results shown:compared with model group, the middle and higher doses of Liuyang Kumquat had markness effect(P<0.05) (except for TG), the middle and higher doses of Yangshuo Kumquat had greatly markness effect(P<0.01) (except for TG). The two middle doses had markness effect to TG(P<0.05), the two higher doses had greatly markness effect to TG(P<0.01).The regulating blood suger effect of kumquat total flavone was studied through evaluating the kumquat total flavone to the diabetes mice’ blood suger, glucose endurance and the normal mice’blood suger. The results shown:compared with model group, after 6 days, the two higher doses had markness effect(P<0.05) to the diabetes mice’ blood suger, after 17 days, the two middle doses had improving glucose endurance, mealwhile, the two higher doses had greatly markness effect (P<0.01) to the diabetes mice’ blood suger. But, all doses had no effect to the normal mice.The effect of kumquat total flavone on the immunity system was studied, through evaluating the phagocytic function of monocyte macrophage, the ear tumefaction induced by DNFB, swimming time, capacity of lacking of Oxygen, the index of thymus and spleen. The results shown:compared with model group, the two middle doses had markness effect(P<0.05), the two higher doses had greatly markness effect(P<0.01).The anti-microbial and security of kumquat total flavone was studied, through evaluating the anti-microbial of kumquat total flavone to E.col and Staphylococcus aureus and maximal tolerance dose test. The results shown:compared with model group, the kumquat total flavone had obviously restrain effect to E.col and Staphylococcus aureus and the maximal tolerance dose was 600 times more than human’ absorbtion.The effect of kumquat total flavone on the gastrointestinal absorption function was stydied, through evaluating the stomach vacuation, small intestine movement rate, small intestine absorption function. The results shown:compared with model group, the two middle doses had markedness effect(P<0.05), the higher doses had greatly markedness effect(P<0.01). This shown that the kumquat total flavone could improve the gastrointestinal absorption function.From the study above, the conclusion was got that the kumquat total flavone had anti-oxidative, moderation hyperlipaemia, regulation blood suger, improvement immunity, anti-microbia, promotion gastrointestinal absorption function, it is a kind of very safe function ingredient and the growth entironment had prodigious effect to the physiology effect.
