枸杞中枸杞色素、枸杞黄酮及枸杞多糖提取工艺的研究

【摘要】 枸杞隶属于茄科(Solanaceae)枸杞族枸杞属(Lycieae L.)植物的果实。随着枸杞药理研究的深入进行,新的药理作用不断被发现.现代医学研究证明:枸杞有免疫调节、抗氧化、抗衰老、抗肿瘤、抗疲劳、降血糖、降血压、补肾、保肝、明目、抗辐射损伤等功效。随着枸杞需求量逐日猛增,使人工种植枸杞日益受到人们重视。对种植枸杞的质量提出了新的要求,本课题选用甘肃白银景泰枸杞为研究对象,研究了枸杞色素的超声波提取工艺;枸杞黄酮和枸杞多糖的微波提取工艺;枸杞黄酮浸膏的分离纯化,建立枸杞植物综合利用的最佳流程。(1)获得了以白银枸杞为原料提取枸杞色素的最佳生产工艺及参数:采用超声波热回流提取技术,提取溶剂为石油醚:氯仿体积比=1:1的溶剂体系;超声波提取功率为:160W;料液比1:8;提取温度55℃;提取时间40min/次,提取次数2次,可实现枸杞浸膏得率3.20%,浸膏中B.胡萝卜素含量达17.5%,枸杞色素的色价值达到129以上的优化提取生产工艺技术。(2)获得了以提取枸杞色素后的枸杞残渣为原料提取枸杞黄酮的最佳生产工艺及参数:采用微波热回流提取技术,提取溶剂为工业乙醇溶剂体系;微波提取功率为:400W;料液比1:12;提取温度80℃;提取时间40min/次,提取次数2次,可实现枸杞黄酮浸膏得率62.72%,浸膏中总黄酮含量达2.69%,枸杞残渣中总黄酮含量达1.69%的优化提取工艺。(3)从甘肃白银景泰枸杞黄酮浸膏中分离得到6种化合物,用化学和波谱学方法鉴定了其中5种化合物的化学结构。它们是β-胡卜素(β-carotene),β-谷甾醇(β-sitosterol),金合欢素(Acacetin),木樨草素(luteolin),芦丁(Rutin)。(4)获得了以枸杞残渣为原料提取枸杞多糖的最佳生产工艺及参数:采用微波热回流提取技术,提取溶剂以水为萃取溶剂;微波提取功率为:500W;料液比1:12;提取温度90℃;提取时间40min/次,提取次数2次,可实现枸杞多糖浸膏得率13.56%,浸膏中总多糖含量达55.35%的优化提取工艺。
【Abstract】 The Lycium chinensis for medicinal purposes part is the fruit of the Solanaceae the (Lycieae L) piant.New pharmacological constantly being discovered,Modern medical studies have shown:wolfberry have immunomodulatory,antioxidant, anti-aging, anti-tumor, anti-fatigue,lowering blood sugar, lowering blood pressure, kidney, liver, eyesight, resistance to radiation damage and other effects.The demand of the Lycium chinense increases suddenly day after day,causes artificial planting medlar to be valued day by day.The people have proposed the new request on the quality of the Planting wolfberry. Wolfberry supplied by Jing tai,Bai yin is the object of study for this topic,experiment in researching pigment ultrasonic-assisted extraction process;Total flavonoids microwave extraction process; Total polysaccharides microwave extraction process;Purification of flavonoids extract Lycium.The experiment establishes the best comprehensive utilization process of the wolfberry.(1)The optimum extraction process of ultrasonic-assisted is selected with the orthogonal design.The yield of pigment were used to evaluate the factor levels.The exttaction process is:(Petroleum ether+chloroform),temperature,55℃,the ratio of solid to liquid,1:8;Ultrasonic power,160w and the return of ultrasonic-assisted 40min; The process of total carotenoids extract,guarantee a highter content,and optimize Lyceum barbarum L total carotenoids the extraction process,Wolfberry color pigments reach to more than the value of 129.(2) The orthogonal design was used in the research on the extraction technique of total flavonoids from LyceumbarbarumL.with organic solvent assisted with microwave.The optimum conditions on the extraction of total flavonoids from Lyceum barbarumL.was as follows:ethanol:96%;the ratio of solid to liquid,1:12; Microwave power 400W;The return of microwave-assisted,40min.Achieve wolfberry extract flavonoids yield 62.72%,total flavonoids from dry extract up to 2.69%.The technique ensured a high content of flavonoids extracted, which was the optimization extraction measure of total flavonoids from LyceumbarbarumL. residue.(3) Six kinds of compounds were isolated from flavonoids extract,in which,five kinds of compounds were identified by chemical and spectral methods,they are tetracosane,β-carotene,β-sitosterol,Acacetin,luteolin,Rutin. (4)The orthogonal design was used in the research on the extraction technique of total polysaccharides from LyceumbarbarumL.with wather assisted with microwave. The optimum conditions on the extraction of total polysaccharides from Lyceum barbarumL.was as follows:wather;The ratio of solid to liquid,l:12;Microwave power 500W;The return of microwave-assisted,40min.Achieve Lyceum barbarumL extract polysaccharides yield 13.56%,total polysaccharides from dry extract up to 55.35%. The technique ensured a high content of polysaccharides extracted, which was the optimization extraction measure of total polysaccharides from LyceumbarbarumL. residue.
