自动IHC染色机控制软件关键技术研究及实现

【摘要】 随着IHC技术在病理诊断和鉴别诊断中的地位不断提高,做好IHC染色工作显得尤为重要。在实际操作与应用中,人工染色易受情绪、时间、环境、方法等影响,就算技术再好也得不到稳定理想的结果。使用自动染色机代替人工染色,实现染色过程的自动化成为当前IHC技术发展的一个趋势。本论文以美国Biocare Medical公司的自动IHC染色机开发项目作为背景,研究自动IHC染色机的工作原理、控制软件及其实现方法。控制软件包括IHC染色规程的制定、抗体与试剂的选择、玻片与试剂的自动定位、染色规程的自动运行与实时监控等功能。论文首先介绍了自动IHC染色系统的组成,分析了系统中各组成部分的作用,研究了控制计算机、运动控制卡、染色机的工作原理和编程接口,给出基本染色动作的软件实现方法;研究了二维条码技术、玻片与试剂识别标签打印技术、识别标签的扫描识别技术、串行通信技术,实现了玻片与试剂的自动定位;在研究染色规程运行规则的基础上,提出解决染色规程运行的分割运行算法和迭代运行算法,并对两种算法作了对比。在研究上述关键技术的基础上,提出控制软件各功能模块的详细实现方法,开发出了自动IHC染色机控制软件。开发的自动IHC染色机控制软件目前已投入测试使用阶段,使用结果表明该控制软件易于操作与控制,能够长时间稳定运行,满足各种IHC染色实验的使用要求。

【Abstract】 With the status of IHC technology in the pathological diagnosis and differential diagnosis improving, it is particularly important to accomplish IHC staining. In practice, artificial staining is vulnerable to be influenced by the mood, time, environment, methods and so on. Even if the staff has the best technology, it is still diffcult to gain the desired results. It has been a trend to use automatic staining which makes the process of IHC staining run automatically to take the place of artificial staining.The paper’s research background is the development project of IHC autostainer which belongs to Biocare Medical in USA. It studies the working principle of IHC autostainer and the realization of its conrol software. The software achieves the establishment of IHC staining protocol, the selection of antibodies and reagents, the locating of slides and reagents, the automatic running of IHC staining protocol, and so on. First of all, the paper introduces the composition of IHC autostainer system, analyzes the role of each component, studies the working principle and programming interface of control computer, motion controller and staining machine, and gives the realization method of staining action in control software. It realizes the automatic positioning of slides and reagents with studying the 2D bacode technology, identification label printing technology, identification label recognition technology and serial communication technology. It proposes the partition running algorithm and iterative algorithm to solve the running of staining protocol with studying operating rules of staining protocol, and compares the two algorithms. It proposes the development method of the control software’s modules in detail, and develops the control software of IHC stainer.At present, the control software of IHC autostainer has been in testing. The results show that the control software is easy to operate and control, runs stably in a long time, and it meets the aquirements of a variety of IHC staining experiments.
