烟草青枯病诱导抗病性研究

【摘要】 烟草青枯病又称烟瘟,其病原为茄青枯假单胞杆状菌(Pseudomonas solanacearum Smith.),发生广,且难以控制,在南方烟区烟草青枯病已成为最主要的烟草病害之一。目前防治烟草青枯病尚未有十分有效的措施。利用诱导剂诱导植物提高抗病性作为一种新的防治病害途径,具有多抗、高抗和安全等优点,是现代植物病害防治的一条重要途径。本试验选用水杨酸、烟草青枯病菌无致病力菌株、茉莉酸甲酯三种物质作为诱导剂,以K326为供试烟草品种,通过对4-6叶期烟草幼苗进行诱导接种烟草青枯菌处理,以接菌当天为Od,连续测定1d、3d、5d、7d、9d、11d、13d烟草幼苗叶片中苯丙氨酸解氨酶(PAL)、过氧化物酶(POD)、多酚氧化酶(PPO)等抗病反应相关的酶活性的变化。并进行盆栽及田间药效对比实验,研究诱导剂在实际生产中的应用效果。以期通过上述研究,提高烟草青枯病综合防治技术水平,进一步丰富和完善诱导抗病性的理论体系。结果表明:1.清水不接种烟草青枯菌处理(CK)组烟草幼苗叶片中PAL、POD和PPO活性在测定过程中一直保持在一个较稳定的水平;清水接菌处理组中烟草幼苗在接菌1-5d叶片中PAL等三种酶活性均较小幅度增加,5d后逐步减少。2.水杨酸、烟草青枯病菌无致病力菌株、茉莉酸甲酯可诱导烟草体内PAL、POD和PPO活性增加,诱导后接种烟草青枯菌可加剧这三种酶变化幅度,其中以1.00 mg/L茉莉酸甲酯诱导接菌处理增加幅度最大。3.盆栽及田间药效试验结果表明水杨酸等诱导剂可诱导烟草增强青枯病抗性。这些结果表明诱导剂处理后的烟草叶片内POD、PAL和PPO活性变化可能与诱导剂提高烟草抗青枯病能力相关。

【Abstract】 Tobacco bacterial wilt is one of main diseases of tobacco in south China due to its wide occurrence and difficultly being controlled, the pathogen is Pseudomonas solanacearum Smith. Today, we can not have a very good mothod to control tobacco bacterial wilt.It is a new way that elicitors induce plants to improve their resistance in controlling disease which has been researched for many years. For its advantage, such as safety, broad spectrum of disease control, it has been paid attention for a long time.In the research, Four-six leaf seedlings of tobacco were used to study the resistance to Tobacco bacterial wilt by Sodium saliculate(SA)、Avirulent bacteriocin producing strains.(ABPS) and Methyl jasmonate(MJ). Determination of PAL、POD and PPO activity in leaves of tobacco seedlings at 1d、3d、5d、7d、9d、11d、13d。Use physiologic and biochemical index to discuss induced resistance mechanism。The results show that SA、ABPS、MJ could increase the activities of POD, PAL and PPO. the activities of POD, PAL Showed a downward trend after the first increase; the activities of PPO is smaller increase in the late.The results suggest that the increased activities of these enzymes is most likely to be associated with the development of inducer resistance in tobacco seedlings 0.3mmol/LSA、108CFU/mlABPS、1.00 mg/LMJ、2.00 mg/L MJ have more obvious changes,1.00 mg/L MJ is the most. The Control effect in the greenhouseand field by different pretreatments shows that 0.3mmol/LSA and 108CFU/ml ABPS have inducing effect against Bacterial Wilt, but not ideal.
