中国荻属植物种质资源遗传多样性的SSR分析

【摘要】 荻属植物,禾本科多年生的C4类高大草本,是一种非常有前途的“能源植物”,近年来在欧美国家受到广泛的关注,正在大力进行开发与利用研究。我国是世界芒荻类植物的分布中心,有着丰富的种质资源,但我国对荻属植物的研究很少,许多品种资源的分类地位及亲缘关系尚不清楚。。本研究应用SSR分子标记技术,探讨荻属植物品种间的遗传多样性。从382对玉米SSR引物和100对甘蔗SSR引物中选出45对条带清晰、并具有多态性的引物,对来自中国黑龙江、吉林、辽宁、山东、江苏、湖南、湖北、宁夏、四川、山西、陕西等11个省的84份荻属及其近缘属植物材料进行SSR扩增。结果表明,45对引物共扩增出248条带,其中多态性条带200个,占80.65%。UPGMA聚类分析结果,84份材料在遗传相似系数GS值0.72水平上,供试验用的84份材料聚成了2个大类,第一大类荻属植物分为4组:第1组,在GS值0.787水平分出,由12份陕西的荻与2份山西的荻组成;第2组在GS0.819水平上分出,7份山西和3份陕西的荻组成;第3、4组在GS0.828水平分开,第3组是由江苏、宁夏、湖南、湖北的荻与南荻组成;第4组是黑龙江、吉林、辽宁、山东的荻和1份江苏的南荻聚在一起。第二大类是5份蔗茅。聚类分析表明荻与南荻遗传相似性很高,聚类呈现较好的地域性分布,且不同来源的荻或南荻没有分别聚为一支,因此认为将南荻作为荻的一个变种处理更为合理。
【Abstract】 Triarrhena. is a perennial potential grass of C4-plant. It is considered as one of the most potential energy plants. At present, it has been used as a raw material of alcohol production and power. In recent year, the European countrys have pay great attentions on the application of Triarrhena. China is the center of Triarrhena of the world. But there is few of research about Triarrhena, its taxonomic treatment has been a controversial matter.By the utilization of SSR markers technique, we discussed the genetic diversity among 79 varieties of Triarrhena and 5 varieties of Erianthus. This study had very theoretical significance and application value for the evaluation, classification, selection of good germplasm resources, utilization and protection. The results showed that: 45 pairs of SSR primer were used to amplify SSR fragments, in order to analyze the genetic diversity among 79 wild samples of Triarrhena in China.45 pairs of SSR primer amplified a total of 248 fragments, of which 200 (80.65%) were polymorphic. Average Nei’s gene diversity is 0.1867 and mean Shannon index is 0.2963. Cluster analysis of UPGMA showed that 84 samples could be clustered into two groups at the level:Erianthus and others.79 samples could be clustered into four subgroups at the level of GD 0.825. The results show that T.lutarioripariaL closely related with T.saccharrflora and genetic diversity of Triarrhena were relation to the areas. We supported the point that T.saccharrflora and T.saccharrflora is a species.
