偶氮染料脱色菌的筛选以及脱色条件优化研究

【摘要】 偶氮染料由于其高色度,成分复杂,这使得其性质稳定,不易为生物所降解,因此对此类废水治理仍然是一个大难题。针对偶氮染料废水的特点,筛选获得了若干具有脱色能力的菌株;并且以氨基黑10B为模型,研究了其脱色的影响因素:包括pH值、O2浓度、添加剂等。首先,从常熟沙家浜染厂边上的塘泥中筛选获得十几株具有脱色能力的菌株,其中1#,18#,25#菌株具有较高的脱色能力,它们在25℃,pH=7条件下,32h之后,其脱色率分别为62.0%、64.12%和71.33%。其次,就这些优势菌株在不同的pH值条件,不同的氧气浓度条件之下,以及在外加添加剂的时候,其脱色的不同变化情况进行了研究,其结果如下:(1)菌株对氨基黑脱色的最佳pH值都是7,1#菌株具有较好的脱色效率,而14#,18#菌株有较广的pH值适应范围。工业生产中选择脱色高效,适应范围更广的菌株,也是非常需要的。(2)经过不同浓度的氧气条件试验,发现在氧气含量较少的时候,存在一个氧气浓度值,在此能达到脱色效率的峰值。(3)当加入L-半胱氨酸时,1#菌株能够在4h之内实现脱色,平时一般需要时间超过28h;KNO3存在情况下,对脱色有抑制作用。
【Abstract】 Among industrial wastewater, azo dye wastewater is one of the most difficult dyes to treat. This is because azo dyes have high colority and complex aromatic molecular structures which make them more stable and difficult to be biodegraded.According to the characteristic of the azo dye wastewater, we screened some decolorizing strains; and we studied the Factors like pH、Oxygen concentration and Additives which control the decolorization with Amido Black 10B as a model.First of all, we got several decolorizing strains which screened from the pool soil beside the dyeing plant in ShaJiaBang. 1#, 18# and 25# strains have more ability to decolorizing the azo dye, with the temperature of 25℃and pH of 7, they made the decolorizing rate of 62.0%、64.12% and 71.33%oSecondly, we studied the factors like pH, Oxygen concentration and Additives which control the decolorizing rate, the following results can be obtained: (1) The optimum pH of the decolorizing strains is 7, strain of 1# has the most ability to decolorizing Amido Black 10B, and strains of 14# and 18# have a wide pH range to decolorizing. The plant need the strains have more effective decolorizing rate and wide pH range. (2) With the experiments of different Oxygen concentration condition, we found that when the Oxygen concentration is very low, there must be a special Oxygen concentration, in which the strains have the most effective decolorizing rate. (3) When L-cysteinyl monohydrochloride was added into the culture medium, the strain of 1# can decolorize the amido black 10B in 4 hours, without it the strain need 28h to decolorizing. On the other side, when we added nitre into the culture medium, the decolorizing rate was reduced.
