黄紫堇化学成分及其抗氧化活性的研究

【摘要】 黄紫堇为罂粟科(Papaveraceae)紫堇属(Corydalis)的多年生草本植物,生于荫的岩旁、溪流两旁、杂木林下或采伐迹地,在我国主要分布在吉林、辽宁、黑龙江等地,具有清热解毒、利尿止痢之功效,主治肺结核、肿瘤、疮毒、咯血等疾病。本文在综述紫堇属植物及黄紫堇研究进展的基础上,对黄紫堇干燥全草的化学成分及生物活性进行了初步探究,主要包括以下几方面的研究工作:1.综述了紫堇属植物化学成分、生物活性以及黄紫堇的研究进展,并且对生物碱含量测定的方法进行了归纳总结。2.对黄紫堇的化学成分进行了预试验,为随后的提取分离工作提供了依据。3.对黄紫堇进行了提取分离。即,将黄紫堇全草醇浸后经酸化有机溶剂萃取,得到了石油醚部分、氯仿部分和正丁醇部分,然后利用柱层析、重结晶、制备薄层色谱等方法对氯仿部分进行分离纯化,得到了两个化合物。依照理化性质、化学方法和1HNMR谱、13CNMR谱、HMBC谱分析初步鉴定了其结构,分别为咖诺定和原阿片碱。4.用GC-MS对黄紫堇石油醚部分的化学成分进行了分离鉴定,分离出29种化学成分,鉴定了其中16种成分并结合归一化法计算了其相对百分含量。5.建立了酸性染料比色法测定黄紫堇氯仿部分总生物碱含量的方法。以溴甲酚绿为染料,盐酸小檗碱为对照品,在415nm波长下测定。经方法学验证可知:该方法简便、准确、可靠,可作为黄紫堇氯仿部分总生物碱的含量测定方法。黄紫堇氯仿部分总生物碱经测定其含量为70.9%。6.对黄紫堇氯仿部分进行了体外抗氧化活性的研究,分别考察了还原能力、清除DPPH自由基、羟自由基、超氧阴离子自由基的能力。结果表明:黄紫堇氯仿部分具有一定的还原能力,并且对以上三种自由基均有不同程度的清除作用。
【Abstract】 Corydalis Ochotensis Turcz is the dry herb of a Papaveraceae perennial herbaceousPlant,grown in the northeast of china.The raw material are used in Chinese folk herbal medicine for treatment of bleeding、ache、tuberculosis、tumor and edema.View from the current study,other medicines of the Corydalis have very significant biological activity in the cardiovascular system, nervous system and so,but for the Corydalis Ochotensis Turcz is still blank in China,so research and exploration its chemical composition and biological activity has great practical significance.In this paper,the chemical compositions and antioxidant activities of Corydalis Ochotensis Turcz were studied. Mainly include the following aspects of the research work:1. After reading a lot of literatures based on the overview of Corydalis plant chemical compositions, biological activities and the research progress of Corydalis Ochotensis Turcz and method for determination of alkaloid content.2. First pre-test the chemical composition of Corydalis Ochotensis Turcz to provide a theoretical basis for the next extraction and separation. The results showed that Corydalis Ochotensis Turcz contains alkaloids, saponins, organic acids, sterols, sugars and glycosides, phenolic compounds, cardiac glycosides. Alkaloid is the main chemical compositions of Corydalis Ochotensis Turcz and content is the highest.3. The dry herb of Corydalis Ochotensis Turcz with 70% ethanol soaking,then acidification and organic solvent extraction,has been part of petroleum ether extract, chloroform extract and n-butanol extract. Use of column chromatography, recrystallization, TLC and other methods chloroform extract of the Corydalis Ochotensis Turcz were isolated and two compounds were isolated. According to physical and chemical properties、chemical methods、1HNMR、13CNMR、HMBC and their structures identified,namely capnoidine (I) and protopine (II). After sincefinder, CA and other database search to identify compound I for the first time I get from the Corydalis Ochotensis Turcz.4. Using the GC-MS the petroleum ether extracts of Corydalis Ochotensis Turcz were studied and 29 kinds of chemical constituents isolated. In NIST1998. L database support and consult relevant literature material identified of 16 kinds of components. Combining normalization method to calculate the relative contents of them. From the results,the main compounds are long-chain diolefine, alkenes, esters, acids. The highest content of long-chain diolefine, the carbon atoms is generally between 14 to 28, mainly have pentacosane (7.02%), octacosane (6.97%)、Tetracosane(6.30%)and the total content of 38.74%. In addition, tricosane, hexacosane, docosane and heneicosane very high content in the total compounds, respectively 5.65%,4.69%,3.57%,2.22%. Hexadecanoic acid(14.67%)、lipids and alkenes content is also high. Can be seen that hexadecanoic acid content is the highest. According to the literature, hexadecanoic acid has anti-tumor activity, this study for the development of antitumor activity provided the basis.5. This is the first established the content determination of chloroform extract of Corydalis Ochotensis Turcz by acid-dye colorimetry.First of all, the determination conditions were screened to determine the best conditions for determination,the Specific means is:With berberine hydrochloride as the control article, precision to measure a certain volume of berberine hydrochloride and test solution in evaporating dish in water bath. Residue with a pH3.6 sodium citrate buffer solution and transferred to a separatory funnel. Bromocresol green solution by adding 7ml, shaking 5min, adding chloroform 10ml, shaking 5min, stand for 1h, chloroform layers obtained by adding 0.5g of sodium sulfate anhydrous, standing for 0.5h, filtered, determination of absorbance at 415nm. Secondly, do the precision experiments、stability experiments、recovery experiments and a series of methodology experiments. Alkaloid content was determined, Its content is 70.9%. The method is simple, reliable, and can be used as content determination method of chloroform extract.6. Studied the Antioxidant activity of the chloroform extract,reducing power and remove ability of the DPPH radical、hydroxyl radical、superoxide anion radical capacity were investigated. The results show that,Chloroform extract has the ability to reducing power and concentration had positive correlation. Chloroform extract can be different levels of three free radical scavenging,remove ability of hydroxyl radicals is weak and remove ability of DPPH radicals and superoxide anion radical is strong.
