近交系小鼠牙胚同种异体移植至口腔粘膜下或牙槽骨内成牙能力的研究
Study on Dentification Ability of Inbred Strain Mice Tooth Germs Implanted into Oral Submucosa and Alveolar Process

【摘要】 目的:观察近交系BALB/C小鼠第一磨牙牙胚移植至成年BALB/C雄鼠口腔粘膜下和(或)牙槽骨内成牙能力,进一步为组织工程牙齿的构建探寻合适的体内生长环境。方法:将出生后4天BALB/C小鼠的下颌第一磨牙牙胚分别植入BALB/C成年雄鼠的口腔粘膜下(右下颌前庭沟)和上颌牙槽骨内(拔除右上颌第一磨牙后,扩大牙槽窝形成),分别于植入后第1、2、3和6周取材,低温脱钙,HE染色,观察牙胚发育的形态学变化。结果:1.牙胚移植至口腔粘膜下后生长良好,牙冠部釉质和牙髓牙本质复合体继续发育,牙本质小管结构清晰,第6周移植物牙冠的形态与正常牙冠的形态相似。第1、2、3周移植物中根方牙本质虽有延长,但仍未能发育成正常牙根的形态。2.牙胚移植至牙槽骨内后仅第1周的移植物制作成切片,移植物内牙本质可见发育,但牙本质小管不清,牙髓及釉质缺失,第2、3、6周移植物回收失败。结论:1.近交系BALB/C雄鼠口腔粘膜下环境有利于BALB/C小鼠牙胚的生长,可为组织工程化牙齿体内模型的构建提供简便合适的发育环境。2.BALB/C小鼠牙胚移植入同种异体BALB/C雄鼠牙槽骨内,牙胚有一定的发育,要使其成为构建组织工程化牙齿的科学模型,还有待进一步改进。
【Abstract】 Objective: To find a suitable environment for the bioengineered teeth in vivo and to investigate the influence of oral environment on the teeth development by observing the dentification ability of BALB/C filial mice tooth germs implanted into BALB/C male mice’s oral submucosa and alveolar process.Method: The first molar tooth germs of the 4 days after birth BALB/C mice were implanted into oral submucosa and alveolar process one by one, and then extract them after 1、2、3 and 6 weeks regularly for histological observation.Result:1. The tooth germs in oral submucosa were growing well with continuing developing enamelum and pulpodentinal complex, and the dentinal tubules were clear. Although the dentine could extend toward the direction of the tooth, the tooth couldn’t develop to be normal form.2. In alveolar process, only the first week implant was made into slice and there was development of dentinum, but the dental pulp was lost, the others were failed to be extracted. Conclusion:1. The environment of the BALB/C male mice’s oral submucosa is favorable for the growth of inbred strain BALB/C mice tooth germs, which can provide a new environment for the bioengineered teeth development in vivo.2. The BALB/C mice tooth germs were developing to some extent when they were implanted into allograft BALB/C male mice’s alveolar process, it needs a further development to be a scientific model.
