益气活血利水法对大鼠大脑中动脉局灶性脑缺血模型(MCAO)治疗作用机制的实验研究

【摘要】 目的:探讨益气活血利水法对局灶性脑缺血大鼠模型神经细胞的保护作用。方法:用线栓法制备大脑中动脉闭塞(MCAO)大鼠模型,为实验对象,分为模型组、假手术组、西药组、中药低剂量组及中药高剂量组。利用化学法、放射免疫分析法及影像学等手段,观察益气活血利水方对MCAO大鼠颅脑CT、中枢神经递质(谷氨酸(Glu)、天门冬氨酸(Asp)、甘氨酸(Gly))、一氧化氮(NO)、内皮素-1(ET-1)、脑钠素(BNP)的改变。结果:益气活血利水方治疗20天,可明显缩小MCAO大鼠颅脑CT中病灶范围大小,改善大鼠神经行为的异常,减少脑组织兴奋性氨基酸(Glu及Asp)的含量、并增加Gly的含量,升高血浆NO、BNP含量,降低血浆ET-1含量。结论:益气活血利水法能显著减少缺血性卒中的神经损伤,改善内皮功能,提高对神经细胞的保护机制,从而起到抗损伤,脑保护的作用。

【Abstract】 Aim: To observe the Yiqihuoxuelishui Recipe on nerve cells of focal cerebral ischemic rats. Methods: Established MCAO rats model with suture for expriment, and the rats were divided into model group, sham-operated group, the group of western medicine,the low-dose and high-dose groups of Chinese medicine. Brain CT, central neurotransmitter (Glu、Asp、Gly), NO, ET-1 ,BNP changes were measured by immunohistochemical method,chemical method,imaging means. Results: After treatment of 20 days, Yiqihuoxuelishui Recipe could significantly reduce the lesion size on brain CT,improve the neurobehavioral abnormalities in rats, reduce the content of brain excitatory amino acids (Glu and Asp),and reduce the content of Gly ,increase serum NO,BNP concentration,reduce the ET-1 expression, obviously improve the pathomorphology changes of nerve cells. Conclusion: Yiqihuoxuelishui Recipe could significantly reduce the nerve injury of ischemic stroke, enhance the protection mechanisms of nerve cells, which play a role of anti-injury and cerebral protection.
