Syk在不同乳腺病变组织中的表达及临床意义

【摘要】 目的:探讨纤维囊性乳腺病、癌前期病变、乳腺癌和乳腺癌旁正常组织中抑癌基因SYK的蛋白表达的差异,并比较Syk的表达与肿瘤大小、淋巴结转移、病理分期、ER、PR、p53和HER2/neu的关系。对象和方法:用SupervionTM二步免疫组化检测20例纤维囊性乳腺病、16例DIN2(导管原位癌,中级别)、168例浸润性导管癌和95例癌旁正常组织中Syk蛋白的表达,并用免疫组化方法检测168例乳腺癌组织中ER、PR、p53、HER2/neu表达的情况。结果:纤维囊性乳腺病、DIN2、乳腺癌及癌旁正常组织中Syk的表达阳性率分别为90%(18/20例)、50%(8/16例)、34.52%(58/168例)、78.95%(75/95例)。乳腺癌与纤维囊性乳腺病(X~2=22.84,P<0.005)、癌旁正常组织(X~2=47.91,P<0.005)中Syk表达率差异有显著意义。DIN2与纤维囊性乳腺病(X~2=7.7887,P<0.010)、癌旁正常组织(X~2=4.646,P<0.05)中Syk表达率差异有显著意义。Syk的表达与肿瘤大小、淋巴结转移、病理分期、ER、PR、p53、HER2/neu均无明显相关性。结论:Syk在纤维囊性乳腺病、癌旁正常组织、癌前期病变和乳腺癌中表达率依次降低。
【Abstract】 ObjectiveTo evaluate the differences of Syk gene expression in human breast fibroadenosis、ductal intraepithelial neoplasia2、breast cancer and adjacent normal tissue,and to study the correlation of expression of Syk gene with tumor size、lymph follicle、stages、ER、PR、p53 and HER2/neu.。MethodsImmunohistochemistry(SupervionTM method ) was used to detect the expression of Syk gene in 20 breast fibroadenosis,16 ductal intraepithelial neoplasia2 and 168 breast cancer(95 adjacent normal tissue),Meanwhile,ER、PR、p53、HER2/neu were detected in 168 breast cancer tissue with immunohistochemical staining.ResultsThe positive rates of Syk gene expression were 90%(18/20)、50%(8/16)、34.52%(58/168)、78.95%(75/95) respectively in breast fibroadenosis、ductal intraepithelial neoplasia2、breast cancer tissue and adjacent normal tissue.The positive expression rate of Syk was significantly lower in the breast cancer tissue than those in the breast fibroadenosis(X~2=22.84,P<0.005) and those in the adjacent normal tissue(X~2=47.91,P<0.005).The positive expression rate of Syk was significantly lower in ductal intraepithelial neoplasia2 than those in the breast fibroadenosis(X~2=7.7887,P<0.010) and those in the adjacent normal tissue of breast cancer(X~2=4.646,P<0.05).We did not find any correlation between reduction of Syk gene expression in breast cancer tissue and clinicopathologic factors or other prognostic markers in our series study.ConclusionProgressive loss of Syk gene expression during progression from normal to DCIS to invasive tumor.No correlation between reduction of Syk gene expression in breast cancer tissue and clinicopathologic factors or other prognostic markers.
