蒙药组方塔米尔对小鼠耐力及抗氧化的影响
Experimental Studies on the Anti-fatigue and Antioxidant Effects of Traditional Mongolian Herbs Tamir in Mice

【摘要】 为开发研究抗疲劳和抗氧化作用的新蒙药,以清洁级昆明种小白鼠为实验动物,对原有蒙药组方那仁满都拉加以改进后的新组方-塔米尔,开展了抗疲劳试验和抗氧化试验。在抗疲劳试验中,将实验动物随机分成4组,Ⅰ组为阴性对照组,灌服等量的羧甲基纤维素(0.5%CM);Ⅱ组为阳性对照组,按0.48g/kg剂量灌服康比特胶囊;Ⅲ组为那仁满都拉组,Ⅳ组为塔米尔组,分别按2.07g /kg剂量分别灌服那仁满都拉和塔米尔散。观察塔米尔散对小鼠负重游泳时间﹑耐常压缺氧时间﹑耐热时间和血乳酸含量,肝糖原和肌糖原含量等的影响。在抗氧化试验中,将实验小鼠随机分成3组,Ⅰ组为阴性对照组,Ⅱ组为那仁满都拉组,Ⅲ组为塔米尔组,给药剂量和途径同上。在末次给药30min后不负重游泳10min,处死取样,测定肝、肾、脑和骨骼肌的总超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-PX)活力,丙二醛(MDA)含量和总抗氧化能力(T-AOC)等。结果显示,蒙药组方塔米尔能显著延长小鼠负重游泳时间﹑耐缺氧存活时间﹑耐热存活时间,显著降低血乳酸含量和显著增加肝及肌糖原含量;蒙药组方塔米尔还能显著或极显著(P<0.05或p<0.01)降低剧烈运动后小鼠肝、肾、脑组织和骨骼肌MDA含量;能显著或极显著(P<0.05或p<0.01)提高脑和骨骼肌组织SOD、CAT、GSH-PX和T-AOC活力,而对肝和肾组织SOD、CAT、GSH-PX和T-AOC活力的影响则无明显的规律性。因此,蒙药组方塔米尔可加速疲劳的消除﹑延缓疲劳的产生,在脑组织和骨骼肌中则呈现较强的抗氧化作用。
【Abstract】 To study and explore the anti-fatigue and antioxidant effects of new preparation of Mongolian traditional medicine-Tamir, the improved preparation based on Narenmandula, the anti-fatigue and anti-oxidation test in mice were carried out respectively in this paper. 48 Kunming mice were randomly divided into 4 groups of 12 in each in anti-fatigue test: groupⅠas a negative control, administered carboxymethyl cellulose(0.5%CM) by per os; groupⅡas a positive control, administered Competitor capsule of 0.48g/kg by per os; groupⅢand groupⅣwas were groups of Narenmandula and Tamir preparation, administered of 2.07g/kg Narenmandula and Tamir by per os, respectively. And then the time of forced swimming, hypoxia survival, heat resisting survival, contents of the blood Lactic Acid and glycogen in mice were observed respectively. 36 Kunming mice were randomly divided into 3 groups of 12 in each in Anti-oxidation experiment: groupⅠas a negative control; groupⅡwas Narenmandula and groupⅢwas Tamir. The liver, kidney, brain and skeletal muscle were sampled immediately finished 10min forced swimming test after 30min of last administratioen, and the activity and content of Total superoxide dismutase(SOD), Catalase(CAT), Glutathione Peroxidase(GSH-PX), total antioxidan tcapacity(T-AOC) and malonaldehyde( MDA ) were determined as soon as possible.The experimental results showed that Mongolian traditional herbs Tamir preparation can significantly prolong the time of forced swimming, hypoxia survival, thermal survival in mice, can decrease significantly the content of lactic acid, significantly increase glycogen content of liver and muscle. Tamir also can significantly decrease the content of MDA in liver, kidney, brain tissue and skeletal muscle, and can enhance the activity of SOD, CAT, GSH-PX and T-AOC in brain tissue and skeletal muscle, however, Tamir has no consentaneous effects on these enzymes activity in liver and kidney. Therefore, Mongolian traditional herbs Tamir preparation can accelerate the elimination of fatigue, can postpone the fatigue and possess strong antioxidant effects in brain tissue and skeletal muscle.
