茶多酚螺旋藻片的研制

【摘要】 本研究课题旨在提供一种适合中老年人食用的抗氧化、增强免疫力的保健品。根据茶多酚本身具有抗氧化作用,螺旋藻本身具有增强免疫力的作用,且螺旋藻中含有的丰富β–胡萝卜素、维生素E与茶多酚具有协同作用[1,2],于是,将二者以一定比例结合起来,研制出茶多酚螺旋藻片,本保健品具有二者的功效,且一定程度上增强了茶多酚的抗氧化作用。首先,进行了茶多酚螺旋藻片的制剂学研究。以崩解度、软材性状情况、软材过筛情况、粘冲情况、片面光洁度作为考察指标,通过单因素实验对辅料进行筛选,优化茶多酚螺旋藻的片芯处方工艺,确定了片芯工艺。最后对片芯进行薄膜包衣,起到一定程度的防潮和防氧化作用。片剂的制备工艺为将处方量的茶多酚、螺旋藻、淀粉、羧甲基淀粉钠、硬脂酸镁混匀,用90%乙醇制成软材,用14目筛制粒后,置80℃干燥2.5小时后,于14目筛整粒,加入羧甲基淀粉钠、硬脂酸镁混匀后,压片,包衣,即得;并制备出一系列样品供质量标准研究和稳定性研究。结果表明该剂型的处方合理、工艺简单。其次,为了控制茶多酚螺旋藻片的质量,对其进行了质量标准的研究。参照《中华人民共和国药典》2005年版对片剂的要求,对片剂的质量进行检查,结果片剂外观完整光洁、色泽均匀,硬度、片重差异、崩解时限均合格;并建立了紫外~可见分光光度法测定片芯中的茶多酚含量、HPLC测定片芯中的表没食子儿茶素(EGC)含量、HPLC测定片芯中的β-胡萝卜素含量的方法,并对方法进行系统考察,结果方法简便、准确、快速、可靠,均可用于该品中成分的含量检测。最后,以外观性状、茶多酚含量、EGC含量、β~胡萝卜素含量、崩解度为指标,进行了茶多酚螺旋藻片的稳定性研究,通过稳定性实验表明:强光、高温对本品均有影响,但是高湿对本品无影响,因此本品应阴凉处保存。

【Abstract】 Based on Tea polyphenol’s antioxidant and Spirulina’s immune enhancement which contains rich spirulinaβ-carotene, vitamin E that have a synergistic reaction with Tea polyphenol, Tea polyphenol Spirulina tablet is prepared by its certain combination. This health-care food is antioxidant and play a role of enhance immune suitable for the middle-aged and elderly and inhance Tea polyphenol’s antioxidant.First of all, the preparation of tea polyphenol and spirulina tablet is studied. Using collapse time, grain’s properties, stiky model properties and tablet’s glabrous’degree as indicator, the formulation preparation technology is optimized by single element experiments. At the end, tablet’s core is coated by a film which can protect tablet from wet and oxidation. The optimal formulation preparation technology are as follows: mixed Tea polyphenols, Spirulina, starch, carboxyl methyl starch sodium and magnesium stearate, then this powder is prepared to be damp mass by ethanol of 90 percent, then granulation, dryness, ground, add carboxyl methyl starch sodium and magnesium stearate, mixed uniformly, pressed and coated. A series of samples are prepared for quality criterion study and stability study. The result showed that the formulation of the dosage form is reasonable, simple.Secondly, the tablet’s quality is determined based on "People’s Republic of China Pharmacopoeia" in 2005 in order to control the quality of tea polyphenol spirulina tablets. As a result, surface is integrity and glabrous, rigidity, weight difference and collapse time are qualified. And the methods of tea polyphenols detected by ultraviolet spectrophotometry, epigallocatechin measured by high performance liquid chromatography andβ-caroten determined by high performance liquid chromatography are established. Then the methods are studied systemly. As a result,they are simple, exact, quick and credibly. They all can be used to determined corresponding components’content.Finally, the tea polyphenols spirulina tablet’s stability is studied. Its experiments show that the tablet is affected by bright, high temperature, but still stability under high humidity. So the tablet should be put in shady and cool.
