SD大鼠视网膜发生发育中Bcl-2及Bax的表达

【摘要】 目的:观察bcl-2及bax在SD大鼠视网膜发育中的表达,探讨二者与视网膜发生、发育的关系。方法:采用免疫组织化学和RT-PCR方法检测胚胎10-20天(E10d-E20d)、生后至成年大鼠视网膜中bcl-2及bax的表达。结果:bcl-2蛋白在胚胎早期几乎没有表达,直至胚胎20天出现表达高峰,继而缓慢下降,在成鼠视网膜中主要于神经节细胞层、神经纤维层和内核层以较低水平维持表达;bax蛋白在胚胎早期就出现阳性表达,缓慢升高,于出生后5天到8天达到高峰,主要表达在神经节细胞层、内核层以及外核层,之后表达迅速减弱,在成年鼠的视网膜组织中几乎见不到bax蛋白的阳性表达出现。结论:Bcl-2、bax在大鼠视网膜胚胎发育中不同时期、不同位置的表达变化,说明大鼠视网膜胚胎发育中的凋亡过程有二者的参与。

【Abstract】 Objective:Overview the expression of bcl-2 and bax in rat retina during development, approach the relationship between the two factors and the development of retina. Methods:Detected the expression of bcl-2 and bax protein in rats’ retina from embryo 10 days to adult by immunohistochemistry and RT-PCR.Results:bcl-2 was not expressed in rat retina during early embryo.The peak of express emerged in the 20th day of embryo,and then decreased gradually.At last, retain with low level in the ganglion cell layer;ganglion-cell layer(GCL),nerve fiber layer(NFL) and inner nuclear layer(INL).The express of bax begin in the early age of embryo,and positive expressed in 5th to 8th day of postnatal,mainly appearance in GCL,INL and ONL.In the retina of adult rat bax almost do not emerge. Conclusion:The different expression of bcl-2、bax in retinals of different-embryo-phase retina rats,which reveal that they take part in the apoptosis program in rat retina during embryonic development.
