P-STAT3和VEGF蛋白在寻常型银屑病患者皮损中的表达及其意义

【摘要】 目的:研究寻常型银屑病患者皮损中P-STAT3和VEGF蛋白的表达及相互关系,并分析它们与PASI评分的关系。方法:采用免疫组化SP法检测35例寻常型银屑病患者皮损组织和16例正常人皮肤组织中P-STAT3和VEGF蛋白的表达,并分析它们与PASI评分的关系。结果:①P-STAT3蛋白在寻常型银屑病患者皮损组织中的表达水平为156.4±11.35,而在正常人皮肤组织中的表达水平为205.6±10.3,两者之间有显著差异(P<0.01);在银屑病进行期皮损组织中的表达水平为142.3±9.11,而在稳定期皮损组织中的表达水平为168.1±9.89,两者相比仍然具有显著差异(P<0.01)。②VEGF蛋白在寻常型银屑病患者皮损组织中的表达水平为151.6±8.21,而在正常人皮肤组织中的表达水平为198.2±9.75,两者之间有显著差异(P<0.01);在银屑病进行期皮损组织中的表达水平为138.4±9.32,而在稳定期皮损组织中的表达水平为159.5±8.11,两者相比仍然具有显著差异(P<0.01)。③P-STAT3和VEGF蛋白在寻常型银屑病患者皮损中的表达呈线性正相关(相关系数为0.74,P<0.01),皮损角质形成细胞中P-STAT3蛋白的表达与PASI评分之间存在线性正相关(相关系数为0.54,P<0.01);而VEGF蛋白的表达与PASI评分之间无线性相关性(相关系数为0.31,P>0.05)。结论:本实验表明:①P-STAT3和VEGF蛋白在寻常型银屑病患者皮损中表达比正常人皮肤组织明显增强,提示它们参与了寻常型银屑病的发病。②P-STAT3蛋白的表达在银屑病进行期比稳定期明显增强,且与PASI评分呈线性正相关,提示P- STAT3不仅与寻常型银屑病的病期有关,而且与疾病的严重程度密切相关。VEGF蛋白的表达在银屑病进行期比稳定期明显增强,但与PASI评分之间无线性相关性,提示VEGF与寻常型银屑病的病期有关,但是与疾病的严重程度无关。③P-STAT3和VEGF蛋白在寻常型银屑病患者皮损中的表达呈线性正相关,提示VEGF基因可能直接由P-STAT3蛋白调节。

【Abstract】 Objective: To study the expression and the significance of P-STAT3 and VEGF proteins in psoriatic skin lesions.Methods: The biopsies were taken from skin lesions in 35 patients with psoriasis vulgaris and the skin of 16 normal controls, the expression and distribution of P-STAT3 and VEGF proteins were detected by immunohistochemical method.Results:①The level of P-STAT3 proteins in lesional skin of psoriasis patients is 156.4±11.35, which is significantly higher than that in normal controls (205.6±10.3, P<0.01). The strong positive expression of P-STAT3 proteins in the active stages of psoriasis vulgaris is 142.3±9.11, but in resting stages is 168.1±9.89. There are significant differences between them (P<0.01).②The level of VEGF proteins in lesional skin of psoriasis patients is 151.6±8.21, which is significantly higher than that in normal controls (198.2±9.75, P<0.01). The strong positive expression of VEGF proteins in the active stages of psoriasis vulgaris is 138.4±9.32, but in resting stages is 159.5±8.11. There are significant differences between them (P<0.01).③There is significant positive correlation between the expression of P-STAT3 and VEGF proteins in the skin lesions (the correlation coefficient is 0.74, P<0.01 ). There is significant positive correlation between the expression of P-STAT3 proteins in psoriatic lesion and PASI index (the correlation coefficient is 0.54, P<0.01), but no correlation between the expression of VEGF proteins in psoriatic lesion and PASI index (the correlation coefficient is 0.31, P>0.05).Conclusion:①The expression of P-STAT3 and VEGF proteins in psoriatic skin lesions are significantly higher than that in normal controls, suggesting that they are involved in the pathogenesis of psoriasis.②The expression of P-STAT3 proteins in the active stage of psoriasis vulgaris is significantly higher than that in the resting stage, and P-STAT3 proteins is positive correlated with PASI index, suggesting that P-STAT3 proteins is not only correlated with the stages of psoriasis vulgaris, but aslo correlated with the severity. The expression of VEGF proteins in the active stage of psoriasis vulgaris is significantly higher than that in the resting stage, but there is no correlation between VEGF proteins and PASI index, suggesting that VEGF proteins is correlated with the stages of psoriasis vulgaris, but is not correlated with the severity.③There is significant positive correlation between the expression of P-STAT3 and VEGF proteins in the psoriatic lesions, suggesting that VEGF gene may be regulated directly by STAT3 protein.
