关于植物组织培养基若干问题的研究

【摘要】 组织培养技术广泛应用于植物科学的许多领域,建立不同植物的组织培养体系是一项非常重要的基础性工作,这里需要研究的问题很多,其中培养基的研究是最为重要工作之一。本文利用多元统计方法对MS等14种较为常用的基本培养基进行了分析,研究了NAA等3种植物激素在母液中的化学稳定性及活性炭对IAA等3种植物激素的吸附作用,取得以下研究结果:1.对MS等14种常用基本培养基多元分析的结果表明:在1.40的聚类水平上可将14种培养基分成4类,第1类包括3种培养基(White,Miller,Heller),第2类包括6种培养基(MS,LS,ER,NT,NN,H),第3类包括3种培养基(B5,N6,SH),第4类包括2种培养基(DKW,WPM):大量元素第1、2主成分分别代表Na2SO4和KNO3,它们权重系数分别为-0.38256和-0.49446。微量元素第1、2主成分分别代表MnSO4.H2O和AICl3,它们权重系数分别为0.41208和-0.47819。有机成分第1主成分代表叶酸和生物素,它们的权重系数均为0.60015,第2主成分代表代表肌醇和硫胺素,它们的权重系数分别是0.53281和0.53052。2.对NAA、IBA和KT 3种植物激素在溶液中化学稳定性的研究结果表明:3种植物激素在水溶液中长期保存均易造成分解,其母液可准确使用的保存时间为不超过10d;温度和光照条件对NAA在水溶液中的稳定性无明显影响,在需要较长时间保存使用时,IBA母液必须遮光保存,KT母液必须低温遮光保存;以95%酒精为溶剂可以大大提高KT、IBA和NAA在母液中的稳定性,对于需要较长时间保存使用的KT、IBA和NAA母液应以95%的酒精为溶剂配制。3.活性炭对IAA、KT和2,4-D 3种植物激素吸附作用的研究结果表明:活性炭对IAA和KT的吸附能力远远大于对2,4-D的吸附能力;不同粒级活性炭对3种激素的吸附能力均差别不大;当震荡时间超过12 h时活性炭对3种激素的吸附量均明显增加;在试验范围内,IAA、KT和2,4-D 3种激素的被吸附量均与活性炭用量呈显著线性相关;MS培养基成分、植物激素及活性炭添加的先后顺序对激素的被吸附量无明显关系。
【Abstract】 Based on the tissue culture system establishment of more and more plant species,the technique of tissue culture is used widely in many fields of plant science.In the process of establishing tissue culture system of a particular plant species,many aspects should be studied,of which,the medium is the most important one.In this paper,14 kind basal mediums were compared and analyzed using multianalysis methods.The chemical stability in different storage conditions of NAA,IBA and KT solution,and the absorption effect of active charcoal for IAA,KT and 2,4-D were studied.The main results are as follows:1.Fourteen kind basal mediums used in plant tissue culture research commonly,were studied using the methods of cluster analysis and principal components analysis.The results showed that:(1) Under the 1.40 classification level according to Chi-square distance,all the 14 kinds basal mediums could be classified into 4 groups.The first group include 3 kinds basal mediums(White,Miller,Heller),the second group include 6 kinds basal mediums (MS,LS,ER,NT,NN,H),the third group include 3 of them(B5,N6,SH),and the fourth group include 2 ones(DKW,WPM);(2) For the macro-elements,the first and the second principal components represent Na2SO4 and KNO3,and there weight coefficient are -0.38256 and -0.49446 respectively,and for the micro-elements,are MnSO4.H2O and AICl3 and 0.41208 and -0.47819 respectively.For the organic substances,the first principal component represent folacin and biotin,and there weight coefficient are all 0.60015,and the second principal component represent myoinositol and thiamine,and there weight coefficient are 0.53281 and 0.53052 respectively.2.The chemical stability in different storage conditions of NAA,IBA and KT solution were studied.The results showed that:(1) In any case tested,the 3 kinds plant growth regulator will decompose gradually in water solution when stored for more than 10 days.So, for the accuracy of experiments,their solution can only be used within 10 days;(2) Temperature and light conditions have no affect on the stability of NAA.But the dark condition can help keep the stability for IBA,and the lower temperature and dark conditions for KT;(3) Compared with water,in 95%alcohol KT,IBA and NAA can keep stability for longer time.Thus,if a long time of storage is need,the solutions of KT,IBA and NAA should be made with 95%alcohol.3.The Absorption effect of active charcoal for IAA,KT and 2,4-D used in tissue culture were studied.The results showed that:(1) The absorption ability of active charcoal for IAA and KT is much larger than for 2,4-D;(2) The absorption ability of different particle diameter of active charcoal for the 3 kinds of plant hormones is almost the same;(3) When the time of absorption longer than 12h,the amounts absorbed of all the 3 kinds plant hormones were increased obviously;(4) In a range studied,the amounts absorbed of all the 3 kinds plant hormones showed a significant linear correlation with the amounts of active charcoal used;(5) The sequence of active charcoal,plant hormones and MS medium ingredient added has no influence on the amounts absorbed of all the 3 kinds plant hormones.
