川牛膝醇提物对SHR大鼠血压及血浆丙二醛、谷胱甘肽含量影响的实验研究

【摘要】 原发性高血压(EH)是一种严重影响人类健康和生活质量的疾病。近年来其发病率呈显著上升趋势,EH所致的心脑血管系统并发症如左室肥厚和脑出血等严重威胁着人类的健康,使患者的生存质量明显下降。由于EH的发病多为遗传、环境、生物等多因素共同决定的,其发病涉及体内物质代谢的紊乱和多系统多器官的病变及功能失调,故单纯降压并不能完全保护和逆转靶器官或组织的损害。因此,寻求有效的中医药治疗途径,发挥中医药的整体调节优势有着十分重要的意义。有关研究已表明,中医药治疗高血压病在改善临床症状,提高生存质量,降低心脑血管系统并发症及靶器官损伤,改善物质代谢等方面,疗效确切,呈现出巨大的发展潜力和广阔的应用前景。高血压病病机极为复杂。牛膝类药材为苋科多年生草本植物,以干燥根入药,是我国重要的大宗药材之一。牛膝在我国有悠久的药用历史,早在汉代就被收载入我国最早的药学专著《神农本草经》,被列为上品。陶弘景谓:“其茎有节似牛膝,故以此为名也。”《本草正义》记载“牛膝疏利泄降,所主皆气血壅滞之病,脑中痛者,多阳邪之上升,牛膝下行为顺,则气火自潜”。利用牛膝引其气血下行,并引其浮越之火下行,可以减轻头部充血,治疗由于肝肾阴亏、肝阳上亢所致头目眩晕、目胀耳鸣、脑中热痛、心中烦热等症状。近年来国内外对牛膝类药材开展了多方面的深入研究,为牛膝类药材的进一步研究提供理论依据。为了进一步阐明牛膝的降压作用及有关机制,予以客观的、科学的论证,我们以自发性高血压大鼠(SHR)为研究对象,使用牛膝为研究药物,进行了以下的研究:1、牛膝对SHR血压的影响;2、牛膝对SHR血浆丙二醛和谷胱甘肽的影响。实验结果显示:1、牛膝能够降低自发性高血压大鼠(SHR)的血压;2、牛膝具有抗氧化作用,能够降低血浆丙二醛(MDA)含量;并增加血浆谷胱甘肽(GSH)含量。通过本课题的研究,从多方面,多层次对牛膝的作用机制进行了探讨,为临床应用牛膝治疗高血压病提供了客观依据。
【Abstract】 Essential hypertension (EH) is a kind of disease that seriously affects the health and living quality of human being. The morbidity of essential hypertension is on the increase in recent years. Some syndromes such as left ventricles hypertrophy and hemorrhage of the brain are threatening the health of people greatly and resulting in the decrease of their living qualities.As EH is dependent on many factors including heredity, environment, biology, and its occurrence is related to the turbulence of physical metabolization, the pathological changes and the functional maladjustment involved in many organs and systems, only lowering the blood pressure could not completely protect the target organs and tissues, and reverse their damages. Therefore, it is important to seek a effective therapy with the assistence of traditional Chinese medicines and exert their integral advantages. Some correponding studies show that traditional Chinese medicines have definitely curative effects as evidenced by improving clinic symptoms and physical metabolization, incresing living qualities, decreasing syndromes of cordis and cerebrovascular systems, and the damage of target organs. As a result, traditional Chinese medicines will present a tremendous development potentials and have a good future.The pathomechanisms of essential hypertension are very complex. Radix is a kind of amaranthaceae perennial herb and is a very important traditional herb. It has a very long history in China and has been collected into "Shennong’s Herbal"—the original pharmaceutical monograph in Han Dynasty. Radix can induce Qi and Xue discending, so relieve cerebral congestion and treat dizzy, tinnitus, thermalgia and sultriness caused by liver and kidney-YIN and excessive rising of liver-YANG. In recent years, all the world have done many researches about Radix and provided many theoretic evidence. To explore the effects and the hypotensive mechanisms of Radix, we used the spontaneous hypertensive rats (SHR) as research object to study the Radix. The observations include the effect on blood pressure, serum malondialdehyde (MDA) level and serum glutathione (GSH) level of SHR..The results show: (1) Radix can decrease blood pressure of SHR; (2) It has antioxidation function via reducing plasma MDA and increasing GSH level of SHR. This research discussed various mechanisms of Radix, provided some objective basis for understanding and using Radix.
