金银花质量控制关键技术与质量评价体系的研究

【摘要】 目的:在中医药学理论和实践的指导下,从金银花本草考证开始,进行系统的资源考察、品种鉴定、质量评价方法与指标、指纹图谱、农残与重金属、以及与金银花药材质量相关的其他研究,寻找并逐步建立更为完善、可靠、科学的中药材质量评价方法,建立金银花质量控制关键技术与质量评价体系。实现中药质量的安全、有效、稳定、可控,实现中药现代化,为中药走向世界奠定良好的基础。方法:结合当前国内外中药质量控制和指纹图谱研究的进展,本课题采用电泳技术和RAPD标记方法对金银花两品系的DNA指纹图谱进行分析,从分子水平上为栽培品种的分类与鉴别提供依据;采用HPLC定性定量分析分别考察不同产地、不同采收期、不同产地加工方法、不同贮藏时间与金银花药材质量的相关性研究;进行金银花药材的常规检查:建立金银花HPLC指纹图谱;构建金银花质量控制规范草案并起草说明。成果与结论:本课题首次采用RAPD标记方法对不同品种金银花的DNA指纹图谱进行了分析,从分子水平上进行了遗传标记研究,为金银花的物种资源描述、评价及鉴定提供了依据。验证了“道地”药材的优势,优选出了最佳采收期、最佳加工方法、最佳贮藏时间,为GAP基地建设提供了数据支撑与技术支撑。建立了全国不同产地的金银花指纹图谱共有峰16个,山东金银花指纹图谱共有峰19个,并且首次在金银花HPLC指纹图谱中标定出绿原酸、咖啡酸、芦丁、木犀草苷、槲皮素等5个已知峰。首次对金银花中六六六(总BHC)、滴滴涕(总DDT)、五氯硝基苯(PCNB)3种农残进行了含量检测分析,为药典金银花农残限量标准的界定提供数据支持,建立了金银花质量控制规范草案。

【Abstract】 Objective:Under the guidance of the theory and practice of Chinese medicine,made a systematic study began with textual criticism of Flos lonicerae and resources,species identification,evaluation methods and quality indicators,fingerprints,pesticide residues and heavy metals,as well as the quality of medicines related to Flos lonicerae.To identify and gradually establish a more complete,reliable,scientific evaluation of the quality of traditional Chinese medicines,establish a key technology of quality control and quality evaluation system for Flos lonicerae.To achieve a safe,effective,stable and controllable quality of Chinese medicine,reach the modernization and lay a good foundation to the world for traditional Chinese medicine.Methods:Connect with the quality control and fingerprints of the progress at home and abroad,the subjects using electrophoresis and RAPD markers to analysis DNA fingerprinting on two strains of Flos lonicerae at the molecular level for the cultivars provide a basis for classification and identification.Qualitative and Quantitative Analysis by HPLC were investigated from different sources and harvest time,processing methods from different areas,different storage time and the quality of Flos lonicerae related studies.Carry out routine examination of Flos lonicerae.Build HPLC fingerprint collection of illustrative plates.Structure the quality control Standard draft and draft explanation of Flos lonicerae.Results and conclusions:This subjects use RAPD markers for different varieties of Flos lonicerae conducted DNA fingerprint analysis for the first time,at the molecular level on the genetic marker studies,and provides a basis for Flos lonicerae species resource description,evaluation and identification.Verify the "Genuineness" ingredients advantages, optimize out the best optimum harvest period,the best processing methods and the best storage time for the construction of GAP data base support and technical support.Set up 16 common peaks of different origin Flos lonicerae fingerprints,and 19 common peaks of Shandong Flos lonicerae fingerprints.And for the first time successfully mark chlorogenic acid,caffeic acid,rutin,luteolin glycosides and quercetin five known peaks in HPLC fingerprint of Flos lonicerae.For the first time detect 666(total BHC),DDT(total DDT), pentachloronitrobenzene(PCNB) for the three kinds of pesticide content,and provide data to support the pesticide limits standards which the Pharmacopoeia defined to Flos lonicerae. Construct the quality evaluation system of Flos lonicerae finally.
