以中药为基质的灰树花发酵工艺条件的研究

【摘要】 灰树花是一种食、药两用真菌,其代谢产物胞外多糖具有抗肿瘤、提高机体免疫力的药理活性。本研究主要是向灰树花液体发酵培养基中添加苦荞、天麻等六种贵州特色中药资源,考察它们对灰树花胞外多糖产量的影响。1、论文研究表明:药用真菌灰树花可在代谢过程中转化中药的有效成分,提高中药活性成分在复合制剂中的比例及效价;此外,中药中的某些成分能够促进药用真菌的生长或活性产物的产生,从而提高灰树花胞外多糖的产量。2、灰树花胞外多糖提取工艺:取100mL发酵液,离心(4000r/min 20min),向上清液中加入95%酒精到乙醇浓度为30%,4℃冰箱过夜,离心(4000r/min,1min),弃去沉淀,上清液继续追加95%酒精到乙醇浓度达75%,4℃冰箱过夜,离心(40000r/min,1min)得沉淀烘干至恒重,即为灰树花胞外多糖。3、灰树花液体发酵培养基:葡萄糖30g/L,蛋白胨5g/L,KH2PO4 4g/L,MgSO4 1g/L;摇瓶发酵培养条件:初始pH自然,接种量10%,种龄5d,温度25℃,250ml三角瓶装液量100ml,转速150r/min。4、在灰树花深层发酵培养基中,当苦荞、天麻、山药的乙醇提液添加量分别为6g/L,6g/L,8g/L时,灰树花胞外多糖产量最高,分别为:3.28g/L、2.31g/L、3.42g/L。5、将苦荞、天麻、山药的乙醇提取液两两组合,添加到灰树花发酵培养基中,得苦荞、天麻组合比例1:2时,胞外多糖产量为3.96g/L。6、应用响应曲面法对试验数据进行分析得出:在灰树花发酵液体培养基中,添加的苦荞乙醇提取液质量浓度为3.0801g/L,天麻乙醇提取液质量浓度为9.4019g/L,山药乙醇提取液质量浓度为7.8856g/L时,灰树花胞外多糖产量最高,达4.6052g/L,较好的解决了灰树花胞外多糖产量较低的问题。
【Abstract】 Grifola frondosa is a kind of fungi both edible and medical,of which the extro-polysaccharide has the functions of anti-tumor and enhance the body immunity. this study was mainly focused on the extro-polysaccharide yield of Grifola frondosa by adding six GuiZhou Local materials,such as Buckwheat、Rhizoma Gastrodiae etc.1、Our study proved that Grifola frondosa could transform the active ingredients in Chinese herb and promote their activities,in addition,the ingredients in Chinese herb could also promote the yield of Grifola frondosa extro-polysaccharide.2、The test results of Grifola frondosa extro-polysaccharide extracting process flow was:take 100mL zymotic fluid,centrifuge in 4000r/min for 20min.then adding ethanol to 30%,keep in 4℃for one night.Centrifuge in 4000r/min for 1min. adding ethanol to the fluid to 75%,keep in 4℃for one night.Centrifuge in 4000r/min for 1min.parch the deposit.3、The Grifola frondosa submerged culture medium composition was:glucose 5g/L, peptone 5g/L,KH2PO4 4g/L,MgSO4 1g/L;and the culture condition was:natural initial pH,seed age 5d,inoculums concentration 10%,250mL triangular flask containing 100mL,and culture under 150r/min and 25℃.4、The best adding quantities of Buckwheat、Rhizoma Gastrodiae、Dioscorea opposite Thunb ethanol extract was 6g/L,6g/L,8g/L.and the Grifola frondosa extro-polysaccharide yield was 3.28g/L,2.31g/L,3.42g/L.5、Take two of Buckwheat、Rhizoma Gastrodiae、Dioscorea opposite Thunb ethanol extract in to consideration,the mix of Buckwhea and Rhizoma Gastrodiae was best,the scale was 1:2,and the yield of extro-polysaccharide could reach 3.96g/L.6、This study optimized the data by responding surface method,the result showed that the yield of Grifola frondosa extropolysaccharide was 4.6052g/L when the ethanol extract quantities of Buckwhea and Rhizoma Gastrodiae was 3.0801g/L, 9.4019g/L.
