贵州香猪实验动物化配套技术研究
Studies on Suitable Technologies of Laboratory Animalization in Guizhou Xiang Pig

【摘要】 贵州香猪属于小型猪,长期近亲交配,遗传稳定,器官结构和功能与人体相似,是十分理想的实验动物。本研究以8世代和9世代贵州香猪为试验对象,研究了剖腹取胎技术、早期断奶技术、超数排卵技术、胚胎移植技术和部分生物学特性。试验结果如下:1.对2头8世代贵州香猪剖腹取胎试验,结果表明:试猪出现阴唇红肿,奶梗明显,最后一对乳头能够挤出奶水等分娩征兆时即可施行剖腹取胎术;速眠新Ⅱ注射液对贵州香猪的全身麻醉效果较好,剂量为0.03 ml/kg,麻醉前20~30分钟肌肉注射硫酸阿托品,剂量为0.04/kg,效果更好;腹中线旁切口手术通路适合于贵州香猪的剖腹取胎手术。2.对39头9世代贵州香猪零天断奶人工哺乳试验,结果表明:零天断奶人工哺乳可以有效地防止疫病的垂直传播,杜绝主要传染病的发生;人工乳配方中用奶粉代替牛乳,并加入5‰的猪血浆蛋白粉,按体重的20~30%进行饲喂,一天饲喂4-6次,能够防止仔猪消化不良,5~7d开始补饲乳猪颗粒料有利于零天断奶仔猪的成活;选择体重较大、活泼健壮的仔猪进行零天断奶人工哺乳试验有利于试验的成功,且公猪比母猪成活率高,初次试验可以选择较多的公猪。3.对35头9世代贵州香猪超数排卵和胚胎移植试验,结果表明:应用PG+PMSG+HCG组合,在发情周期的14-17天注射PMSG,剂量15-18IU/kg,前一天每猪注射PG0.2mg,注射PMSG后72h注射等量的HCG,超排效果较好;将31枚可用胚胎移植给同期发情的4头受体,有2头受孕,最后1头产仔。4.通过开展贵州香猪部分生物学特性研究,结果发现贵州香猪的发情周期比普通猪种少1天,平均为20天,并且在哺乳期间便有发情现象,如果配种,可以正常妊娠;6月龄贵州香猪33项血液生理生化指标中,有17项指标在人类血液生理生化指标正常范围之内。5.本课题的研究为贵州香猪的净化提供了技术平台和重要的参考数据,为实现贵州香猪的实验动物化奠定了基础,填补了这方面研究的空白。通过本研究,为贵州香猪的净化提供了技术准备及重要的参考数据,从而为实现贵州香猪的实验动物化奠定了基础。
【Abstract】 Guizhou Xiang pig belongs to the Miniature pig. Because of long-term inbreeding, the heredity is stable, and the organ function and the organizational structure are similar to the human body. So it is the extremely ideal experimental animal. This topic has studied the techniques of Caesarean section, Embryo transplantation, Early weaning, superovulation and some biological characteristics through the eighth generation and ninth generation Guizhou Xiang pig. The result is as follows:1. The result of Caesarean section to two eighth generation Guizhou Xiang pigs indicated: It can perform the operation of Caesarean section when the Guizhou Xiang pigs appear the symptom of the red swelling of the labium skin, extruding milk of the last nipple. Su Mian XinⅡis good to Guizhou Xiang pig’s general anesthesia, the dosage is 0.03 ml/kg. The effect will be better to inject atropine sulphate before 20~30 minute, the dosage is 0.04/kg. The lateral incision of medioventral line is fit to the Caesarean section of Guizhou Xiang pig.2. The result of artificial nursing experiment of zero-day weaned 39 ninth generation Guizhou Xiang pigs indicated: Under the isolated condition, artificial feeding on zero-day could prevent loemic vertical transmission, eliminate the occurrence of main communicable disease. It can prevent swine’s dyspepsia to take the place of cow’s milk by milk powder in the prescription of artificial milk, and add in 5‰plasma protein powder, and feed 4-6 times one day. The first experiment could choose some piglets of bigger body weight and more boars.3. The results of superovulation and embryo transplantation experiments to 35 ninth generation Guizhou Xiang pigs indicated: The superovulation effect is good using the PG+PMSG+HCG combination. It can inject PMSG on oestrous cycle’s 14-17 days, each pig is injected PG 0.2mg on the preceding day of, and injected partes aequales’s HCG after 72h of injecting PMSG. The dosage is 15-18IU/kg. 31 available embryoes were transplant to 4 acceptors of synchronization of estrus with donator. 2 acceptors become pregnant, and 1 gives birth to 2 piglets.4. Through carrying out the research of Guizhou Xiang pig’s some biological characteristics, the result indicated: Guizhou Xiang pig’s oestrous cycle is 20 days. It is 1 day less than the ordinary pig. It has the phenomenon of empathema in the period of lactation, and it can be pregnancy if being bred. In June in the age Among the 33 targets of blood physiology biochemistry of 6 Guizhou Xiang pigs in the age of 6 months, it has 17 blood physiology biochemistry targets within the normal human blood physiology biochemistry target.5. It has laid the foundation for Guizhou Xiang pig’s purification. It has also provided the technology platform and the important reference data to realize its laboratory animalization. It has filled the blank of this aspect’s study.This study has provided the technical preparation and the important reference data for the purification of Guizhou Xiang pig, accordingly lain the foundation to realize Guizhou Xiang pig’s experiment animalization.
