微卫星标记多态性与波尔山羊生长性状的相关性研究

【摘要】 本试验根据山羊遗传连锁图谱选择20个微卫星标记,利用PCR技术对波尔山羊基因组DNA进行扩增,用以检测其群体遗传特征并进行了微卫星标记和出生体重、体长、体高、胸围、断奶体重、体长、体高、胸围和周岁体重、体长、体高、胸围的相关性分析,对微卫星标记各位点不同基因型个体间进行了多重比较,分析了各基因型对生长性状的影响。结果表明:所选择的20个微卫星标记在波尔山羊群体中表现出较好的多态性,共检测到193个等位基因,平均在每个位点检测到9.65个等位基因,平均有效等位基因数(Ne)为7.3287,平均多态信息含量(PIC)为0.8290,平均杂合度(H)为0.8478。利用SAS程序下的GLM过程,对20个微卫星位点进行生长性状的最小二乘分析,结果如下:(1)体重方面:INRA011位点228/228、INRA040位点238/238与246/216、ILSTS034位点193/167为出生体重的有利基因型。BM1312位点145/121、INRA011位点240/240、MAF064位点159/159、IDVGA64位点281/243、LSSCV37位点159/143与161/147、BMS1248位点162/162、CSSM047位点153/153、INRA129位点198/172为出生体重的不利基因型。INRA011位点228/228、IDVGA64位点245/245、INRA040位点238/238、ILSTS034位点193/167、BM0415位点136/136为断奶体重的有利基因型。MAF064位点141/141、INRA040位点238/238、CSSM047位点171/171为周岁体重的有利基因型。CSSM047位点171/153、SRCRSP10位点311/275为周岁体重的不利基因型。(2)体长方面:BM1312位点145/117、INRA040位点238/238为出生体长的有利基因型。INRA011位点240/240、MAF064位点159/159、IDVGA64位点281/243、NRA129位点198/172为出生体长的不利基因型。MAF070位点198/184为断奶体长的有利基因型。BM0415位点136/136、MCM527位点203/203与207/207为断奶体长的不利基因型。CSSM011位点206/206、NRA040位点238/238、CSSM047位点171/171、SRCRSP10位点303/269为周岁体长的有利基因型。(3)体高方面:INRA040位点238/238为出生体高的有利基因型。BM1312位点145/121、MAF064位点159/159、IDVGA64位点281/243、LSSCV37位点159/143与161/147、BMS1248位点162/162与142/142、INRA129位点198/172为出生体高的不利基因型。(4)胸围方面:INRA040位点238/238与246/216、INRA129位点178/178为出生胸围的有利基因型,INRA129位点198/172、LSSCV37位点161/147与159/143、MAF064位点159/159、IDVGA64位点281/243为出生胸围的不利基因型。IDVGA64位点245/245为断奶胸围的有利基因型。MAF070位点198/184与198/198为断奶胸围的不利基因型。BM1312位点145/117、INRA129位点178/178为周岁胸围的有利基因型。研究结果将为开展波尔山羊标记辅助选择提供依据。

【Abstract】 Twenty microsatellite loci were selected from the goat linkage map in this study.The microsatellite DNA was amplified using PCR technique to examine the genetic structure and relationship between microsatellite DNA and growth traits including body weight, length,height,chest of birth,weaning and one-year-old.Twenty microsatellite loci analyzed were polymorphic in Boer goat population with all the observed number of alleles,the average effective number of alleles(Ne),an average number of alleles per locus,the average information content(PIC) and average heterozygosis(H) were 193,7.3287,9.65,0.8290 and 0.8478 respectively.Making use of the PROC GLM of SAS we get the result as the follows:(1) 228/228 of INRA011,238/238 and 246/216 of INRA040,193/167 of ILSTS034 were beneficial genotypes to birth weight.145/121 of BM1312,240/240 of INRA011,159/159 of MAF064,281/243 of IDVGA64,159/143 and 161/147 of LSSCV37,162/162 of BMS1248, 153/153 of CSSM047,198/172 of INRA129 were disadvantageous genotypes to birth weight.245/245 of IDVGA64,238/238 of INRA040,193/167 of ILSTS034,136/136 of BM0415 were beneficial genotypes to weaning weight.141/141 of MAF064,238/238 of INRA040,171/171 of CSSM047 were beneficial genotypes to one-year-old weight, 171/153 of CSSM047,311/275 of SRCRSP10 were disadvantageous genotypes to one-year-old weigh.(2) 145/117 of BM1312,238/238 of INRA040 were beneficial genotypes to birth length.240/240 of INRAO11,159/159 of MAF064,281/243 of IDVGA64,198/172 of NRA129 were disadvantageous genotypes to birth length.198/184 of MAF070 were beneficial genotypes to weaning length,136/136 of BM0415,203/203 and 207/207 of MCM527 were disadvantageous genotypes to weaning length.206/206 of CSSM011,238/238 of INRA040,171/171 of CSSM047,303/269 of SRCRSP10 were beneficial genotypes to one-year-old length.(3) 238/238 of INRA040 were beneficial genotypes to birth high.145/121 of BM1312,159/159 of MAF064,281/243 of IDVGA64, 159/143 and 161/147 of LSSCV37,162/162 and 142/142 of BMS1248,198/172 of INRA129 were disadvantageous genotypes to birth high.(4) 238/238 and 246/216 of INRA040,178/178 of INRA129 were beneficial genotypes to birth chest.198/172 of NRA129,161/147 and 159/143 of LSSCV37,159/159 of MAF064,281/243 of IDVGA64 were disadvantageous genotypes to birth chest.245/245 of IDVGA64 were beneficial genotypes to weaning chest,198/184 and 198/198 of MAF070 were disadvantageous genotypes to weaning chest.145/117 of BM1312 were beneficial genotypes to one-year-old chest.This study has a great significance because that it can give a theory gist for genetic marker assistant selection(MAS),Quantitative traits locus(QTL)locating, improvement of Boer goat breeding.
