蜈蚣败毒饮对雌激素周期小鼠阴道上皮细胞有丝分裂的影响

【摘要】 目的:蜈蚣败毒饮对雌激素周期小鼠阴道上皮细胞有丝分裂的影响。方法:取健康昆明雌性小鼠50只,连续3天腹腔注射己烯雌酚,诱导小鼠处于雌激素周期。随机平均分成5组,即蜈蚣败毒饮低剂量组(简称中低组)、蜈蚣败毒饮中剂量组(简称中中组)、蜈蚣败毒饮高剂量组(简称中高组)、复方青黛胶囊组(简称阳性对照组)和生理盐水组。中低组、中中组、中高组分别给予低浓度蜈蚣败毒饮(8.78 g/kg·d)、中浓度蜈蚣败毒饮(17.55g/kg·d)、高浓度蜈蚣败毒饮(35.10 g/kg·d)灌胃,对照组给予复方青黛胶囊悬浊液(0.78 mg/kg·d)灌胃,生理盐水组给予生理盐水(15 ml/kg·d)灌胃。于8:00开始给药,连续7天。基于动物细胞有丝分裂的昼夜节律性,每组实验时间均加以统一。第8天上午10:00给5组动物腹腔注射秋水仙碱15 mg/k g,使细胞有丝分裂周期停滞于有丝分裂中期,便于计数。杀死动物,切取阴道组织,制成病理切片。在光学显微镜下观察并计数基底细胞中所出现的有丝分裂数。结果:结果表明,生理盐水组基底细胞中有较多的有丝分裂细胞,细胞核增大,深染,有的分叶。阳性对照组和中药各剂量组有丝分裂细胞均有所减少,阳性对照组、中低组、中中组、中高组、与生理盐水组相比P<0.01;中高组与阳性对照组相比P<0.05。结论:蜈蚣败毒饮能抑制小鼠阴道上皮细胞有丝分裂。蜈蚣败毒饮优于复方青黛胶囊。

【Abstract】 Objective To observe the affection of WUGONGBAIDUYIN decoction towards the mitosis of vaginal epithelial cells of mouse in the estrogen period.Methods 50 KM mouse,female,with intraperitoneal injection of Diethylstilbestrol for3 days,are induced in estrogen period.They are divided into 5 groups randomly. Group 1 is treated with WGBDY decoction( 8.78g/kgd), Group 2 is treated with WGBDY decoction(17.5 5 g/kg·d) ,Group 3 is treated with WGBDY decoction ( 35.10g/kg·d) ,Group4 is treated with suspension of FUFANGQINGDAI capsule, Group 5 is treated with NS.All the medicine are given by gavage,at 8Am,for 7 days.On the 8th day,at 10 Am, all the 5 groups are treated with Colchicine by intraperitoneal injection 15mg/kg, to make the mitosis stagnated in the Medium-term for count.Kill the mouse,cut the vaginal organizations,and make the pathological section.Observe the cells in mitosis among the basal cells with optical microscope and count.Results In group 5,there are more cells in mitosis. Nucleus increase and deep.In other groups,the amount of cells in mitosis decrease.When group 1,2,3,4 are compared with group 5, P<0.01.When group 3 is compared with group 4, P<0.05.Conclusion WGBDY decoction inhibit the mitosis of vaginal epithelial cells of mouse.
