鱼腥草注射液指纹图谱与抗炎、抗菌药效组分的研究

【摘要】 在完善中药材质量标准体系过程中,指纹图谱技术成为鉴别中药真实性及评价质量一致性和产品质量稳定性的实际可行模式。指纹图谱质量控制模式的提出使得中药(材)的研究方法和质量分析手段也由针对一个或者少数几个活性成分(指标成分)的分析,发展成为对整味中药化学成分的综合分析。本论文针对中药注射剂色谱指纹图谱研究中的难点问题:指纹图谱和药效相关性,利用联用色谱仪器,结合化学计量学方法,首次开展了鱼腥草注射液抗炎药效、抗菌活性的实验研究。采用层级聚类分析对不同厂家注射液的指纹图谱进行分类。然后采用二甲苯所致小鼠耳肿胀炎症模型和角叉菜胶所致大鼠胸膜炎模型对抗炎药效进行研究,采用活菌计数法对体外抗菌活性进行测定。结果表明第一类厂家的具有显著的抗炎、抗菌效果,而第二类厂家的抗炎效果并未达到显著性水平。分析表明鱼腥草注射液主要的药效成分为甲基正壬酮、癸酰乙醛、月桂醛、辛醛、β-蒎烯、β-芳樟醇、1-壬醇、4-松油醇、α-松油醇、乙酸龙脑酯、癸酸和乙酸香叶酯等。在体内抗炎和体外抗菌药效基础上建立了鱼腥草注射液的指纹图谱共有模式。为鱼腥草注射液的指纹图谱标准建立提供了有力的支撑,为提高鱼腥草注射液的质量提供了理论和实验数据。

【Abstract】 In the process of improving quality appraisal system for traditional Chinese medicine(TCM),fingerprint technique turns to be a practical and feasible mode to maintain the stability of product quality.And it can better distinguish the authenticity of TCM.The brought up of fingerprint technique promotes a development in research and quality analyzing method from the study for one or several active ingredients to a comprehensive study for the whole TCM.This thesis sets out to settle the problem of chromatographic fingerprint research of TCM injection:the following work was done with hyphenated chromatographic instruments and chemometric approaches:the study of the correlation between fingerprint and anti-inflammatory effect and inhibitory activity.The first attempt on fingerprint-efficacy of Houttuynia cordata injection was developed.The fingerprints of Houttuynia cordata injection from different factories were classified by hierarchical clustering. Anti-inflammatory effect of Houttuynia cordata injection was characterized by the rat pleurisy model induced by carrageenin and the mice ear edema model by xylene.Viable count method was used to determine inhibitory activity of Houttuynia cordata injection. Anti-inflammatory effect and inhibitory activity of the injection from the first class factories on the two models were significant,while the second class were not.The analysis result indicated that main effect compounds in Houttuynia cordata injection are methyl n-nonyl ketone, decanoylacetaldehyde,lauryl aldehyde,capryl aldehyde,β-pinene,β-1inalool,1-nonanol,4-terpineol,α-terpineol,bornyl acetate,n-decanoic acid and acetic acid,geraniol ester etc.The author then established a corresponding relationship between the fingerprint of Houttuynia cordata injection and efficacy to certain extent. The pattern of Houttuynia cordata injection is based on the efficacy.The mode established in this thesis provides specific experiment data and also a solid foundation for the establishment of Houttuynia cordata fingerprint standards.
