再生柞蚕丝丝素静电纺及结构研究

【摘要】 本文研究了再生柞蚕丝丝素蛋白的静电纺参数对纤维平均直径和结构的影响,静电纺纤维中溶剂六氟异丙醇(HFIP)的残留量及乙醇后处理对非织造网结构的影响和细胞对柞蚕丝丝素蛋白静电纺非织造网的相容性。预实验寻找可以进行静电纺的影响因素中溶液质量分数、电压及接收距离的范围。正交试验分析这三个可控制因素对纤维平均直径的影响程度,依次为质量分数、电压、接收距离。用X-射线衍射法和热分析法分析不同参数组合对非织造网结构的影响,发现静电纺丝有使结构从α-螺旋或无规卷曲向β-折叠转变的趋势,但纺丝参数在可纺范围内变化对构象的转变影响较小,仅依靠改变参数促使非织造网结构向更稳定的β-折叠状态的作用很小,需要通过其它的后处理手段才能实现。用HFIP作溶剂进行静电纺丝后,纤维中会残留溶剂,用色谱-质谱法测定HFIP在未处理和用乙醇处理后纤维中的残留,发现未处理纤维中溶剂残留较大,用纯乙醇处理后再用85%乙醇处理可以有效降低残留量。但乙醇处理会引起非织造网收缩、溶解,用扫描电镜观察乙醇处理后纤维表面形态的变化,用红外光谱和X-射线衍射法比较处理后二级构象的变化。在非织造网上培养小鼠成纤维细胞,用显微镜观察细胞在材料上的粘附、生长和增殖情况,采用四唑盐(MTT)比色法细胞增殖试验测定细胞在材料上的增殖情况,研究细胞与非织造网的相容性,发现细胞可以在纤维上黏附、生长,但纤维在培养液中出现溶解,影响了细胞的生长。
【Abstract】 This paper have studied the effect of the electrospinning parameters of regenerated tussah silk fibroin to fiber average diameter and structure, the residual of solvent (HFIP, 2-propanol,1,1,3,3,3-hexaluoro-methanol ) in fibers and the influence on electrospinning non-woven net after treatment of ethanol,also studied the compatiblity of cell and regenerated tussah silk fibroin electrospinning non-woven net.By preliminary experiment,the scope of solution concentration,voltage and spinning distance which are influencing factors of electrospinning have been obtained. The influencing degree of this three controlling elelments have been analysised by orthogonal experiment. By X-ray diffraction method and thermal analysis sytem, the change of electrospinning non-woven net structure by different parameter have been researched to find the tendency of transition fromα-helix or random coiltoβ-sheet, but the impact to transition of conformation was little by spinning parameters in spinnability scope, so the role to make non-woven net structure more stable is small only by this way, and this aim can come true by other post processiong. After electrospinninging with HFIP as solvent, HFIP can remain in fibers. The residual in undisposed fibers and disposed fiebers by alcohol were elvaluated by GC\MS, and the result is that residual in undisposed fibers is a little large and the residual in the disposed fibers with pure alcohol firstly then 85% alcohol treatment stepped down to zero. But treatment with ethanol can cause shrinkage and dissolution of net. The change of fiber morphology was observed by SEM and the change of second order by infrared spectrum and X-ray diffraction. L-cells were cultivated on non-woven net, and the adhesion, growth and differentiation were observed by microscop. The proliferation of cells on net were determined by MTT to study the compatibility of cell and net. The result showed cells could adhere, grow on net, but because of the dissoluton of fibers in culture solution, the growth of cells have been impaceted.
