西藏胡黄连化学成分的研究

【摘要】 西藏胡黄连(Picrorhiza scrophulariiflora Pennell)为玄参科(Scrophulariaceae)多年生草本植物,药用部位为根茎,主产于西藏、云南、四川等高海拔(4400 m以上)地区,味苦、性寒,为清虚热之要药,始载于《开宝本草》,具有清热凉血、燥湿消疳的功效,主治骨蒸潮热、小儿疳疾、湿热泻痢、黄疸、吐血、衄血、目赤肿痛、疮疡肿毒等症。为明确该中药材中的化学成分,阐明其物质基础,并为后继开发研究提供理论依据,本文利用硅胶、ODS、凝胶柱色谱等分离手段对西藏胡黄连的95%乙醇(V/V)提物的乙酸乙酯萃取物及正丁醇萃取物的大孔树脂50%乙醇/水洗脱部分进行了化学成分分离鉴定,从中分离得到了34个化合物,通过理化性质、化学反应以及波谱分析(NMR),鉴定了其中28个化合物的结构,分别为:正二十六烷醇(1)、β-谷甾醇(2)、胡萝卜苷(3)、25-乙酰氧基-2-β-D-吡喃葡萄糖氧基-3,16,20-三羟基-19-去甲羊毛甾烷-5,23-二烯-22-酮(4)、2-β-D-吡喃葡萄糖氧基-3,16,20,25-四羟基-19-去甲羊毛甾烷-5,23-二烯-22-酮(5)、香草酸(6)、没食子酸(7)、熊果苷(8)、草夹竹桃苷(9)、藏黄连苷G(10)、藏黄连酚苷A(11)、儿茶素(12)、木犀草素(13)、木犀草素-7-O-β-D-吡喃葡萄糖苷(14)、11-O-(4′-甲氧基没食子酰基)-岩白菜素(15)、11-O-没食子酰基岩白菜素(16)、岩白菜素(17)、米内苷(18)、胡黄连苷Ⅰ(19)、胡黄连苷Ⅱ(20)、獐芽菜苷(21)、龙胆苦苷(22)、胡黄连苷Ⅴ(23)、异阿魏酸(24)、咖啡酸-4-O-β-D-吡喃葡萄糖苷(25)、咖啡酸甲酯-4-O-β-D-吡喃葡萄糖苷(26)、1,6-二咖啡酰基-O-β-D-吡喃葡萄糖苷(27)、藏黄连咖啡酸苷A(28)。其中化合物1、7、8、12~17、21、22、24~27为首次在胡黄连属植物中分离得到,化合物23和28为两个新化合物。

【Abstract】 The plant Picrorhiza scrophulariiflora Pennell(Scrophulariaceae)grows in high-altitude regions(above 4400 m)in southeast Tibet and northwest Yunnan province in China.The root of this plant is used in traditional Chinese medicine for the treatment of damp-heat dysentery,jaundice,indigestion,common fever,acute viral hepatitis,and steaming of bone.For the purpose of clear the material base for its bioactivity and provides the theory basis for the successor development research on the root of Picrorhiza scrophulariiflora Pennell,the chemical constituents investigation on the acetic ether layer and n-butanol layer of the 95%ethanol extracts were carried out.By using silica gel C.C.,Sephadex LH-20 and ODS, 34 compounds were isolated,morever,the structures of 28 compounds were identified by the physical-chemical properties and spectral methods,which were identified as hexacosanol(1),β-sitosterol(2),daucosterol(3),25-acetoxy-2-β-D-glucopyranosyloxy-3,16,20-trihydroxy-9-methyl-19-norlanosta-5, 23-diene-22-one(4),2-β-D-glucopyranosyloxy-3,16,20,25-tetrahydroxy-9-methyl-19-norlanosta-5, 23-diene-22-one(5),vanillic acid(6),gallic acid(7), arbutin(8),androsin(9),scroside G(10),scrophenoside A(11),catechin(12),luteolin(13), luteolin-7-O-β-D-glucopyranoside(14),11-O-(4’-O-methylgalloyl)-bergenin(15),ll-O-galloylbergenin (16),bergenin(17),minecoside(18),picrosideⅠ(19),picrosideⅡ(20), sweroside(21),gentiopicroside(22),picrosideⅤ(23),isoferulic acid(24),caffeic acid-4-O-β-D-glucopyranoside(25),methyl caffeate-4-O-β-D-glucopyranoside(26),1, 6-dicaffeoyl-O-β-D-glucopyranoside(27),scrocaffeside A(28).Among them compound 1,7, 8,12-17,21,22,24-27 were isolated from Picrorhiza genus for the first time,compounds 23 and 28 were novel compounds.
