金樱子的化学成分研究

【摘要】 中药金樱子为蔷薇科蔷薇属植物金樱子(Rosa laevigata Michx.)的果实,始载于《蜀本草》,又名刺榆子,刺梨子,金罂子,山石榴,糖罐子等。分布于华东,中南,西南等地。《本草纲目》中记载金樱子:性酸、涩、平、无毒;主治脾泻下痢、止小便利、涩精气。金樱子中含有大量三萜类化合物,本论文对其体积分数70%乙醇提取物进行了分离和结构鉴定,利用硅胶柱色谱,大孔树脂柱色谱,ODS柱色谱,Sephadex LH-20凝胶过滤,制备高效液相和重结晶等分离手段,从中分离得到18个化合物,通过1HNMR,13C-NMR,DEPT,ESI-MS等波谱和光谱学方法鉴定了18个化合物的结构,分别为:乌苏酸(1),坡模酸(2),蔷薇酸(3),1α,2α,3β,19α-四羟基乌苏酸(4),19α-羟基亚细亚酸(5),2α,3α,19α,23-四羟基-12-烯-28-乌苏酸(6),齐墩果酸(7),山楂酸(8),2α,3α-二羟基齐墩果酸(9),2α,3β,19α-三羟基齐墩果酸(10),2α,3β-二羟基羽扇-20(29)-烯-28-酸(11),2α,3β,23-三羟基羽扇-20(29)-烯-28-酸(12),19α-羟基亚细亚-28-O-β-D-吡喃葡萄糖苷(金樱子皂苷A)(13),委陵菜酸-28-O-β-D-吡喃葡萄糖苷(14),β-谷甾醇(15),胡萝卜苷(16),翻白叶苷A(17),山奈酚(18)。其中,化合物2,4,8,9,10,11,12为首次从该植物中分离得到。
【Abstract】 Rosa laevigata Michx.is the fruit of Rosa laevigata Michx.in Rosaceae,it was recorded in "Shu Materia Medica"at first,and has some other names such as Ci Yu Zi,Ci Li Zi,Jin Yao Zi,Shan Shi Liu and Tang Guan Zi.It was widely distributed in the east,middle-south and southwest of China[1].As a traditional chinese medicine Rosa laevigata Michx.was recorded in "Compendium of Materia Medica" to treat dysentery,piss convenience,Shibuya fine gas.[2]By means of column chromatographic Silica gel,Diaion HP-20,ODS,Sephadex LH-20, HPLC and so on,the investigation on the chemical constituents of the 70%ethanol extracts of Rosa laevigata Michx.has resulted in the isolation and structure elucidation of eighteen compounds named Ursolic acid(1),Pomolic acid(2),Euscaphic acid(3),1α,2α,3β,19α-tetrahydroxyurs-12-en-28-oic acid(4),19α-hydroxyasiatic acid(5),Myrianthic acid(6),Oleanolic acid(7),Maslinic acid(8),2α,3α-dihydroxyolean-12-en-28-oic acid(9),2α,3β,19α-trihydroxyolean-12-en-28-oic acid(10),2α,3β-dihydroxylup-20(29)-en-28-oic acid(11),2α,3β, 23-trihydroxylup-20(29)-en-28-oicacid(12),19α-hydroxyasiaticacid-28-O-β-D-glucopyrannoside(13), Tormentic acid-28-O-β-D-glucopyrannoside(14),β-sitosterol(15),Daucosterol (16),PotengriffiosideA(17),Kaempferol(18).Among them,compound 2,4,8,9,10,11,12were isolated from this plant for the first time.
