空肠弯曲杆菌感染分布研究

【摘要】 本研究对入沪二十八个产地(河南开封、山东青岛、江苏南通、浙江金华、上海奉贤等地)的鸡、猪进行了空肠弯曲杆菌流行分布调查,采取“分散选点、等比抽样”的原则,采集了6702(猪3657份、鸡3045份)份肝脏、盲肠内容物、泄殖腔拭子(粪便)样品,采用以VS1基因为目标片段,只有对空肠弯曲杆菌才能扩增出特异性片段,而其它弯曲菌和其它属的细菌都不能扩增出特异性片段的快速、敏感、特异的PCR方法进行检测。调查和研究结果表明,二十八个产地的鸡、猪均有空肠弯曲杆菌感染,不同产地、种属其感染率和发病情况不同。用PCR方法对3045份来自不同产地的鸡肝脏(1015)、盲肠内容物(1015)、泄殖腔拭子(1015)样品进行检测,结果显示肝脏平均阳性率为13.2%,盲肠内容物平均阳性率为28.4%,泄殖腔平均阳性率为19.8%;对3657份来自不同产地的生猪肝脏(1219)、盲肠内容物(1219)、粪便(1219)样品进行检测,结果显示肝脏平均阳性率为10.3%,盲肠内容物平均阳性率为34.4%,泄殖腔平均阳性率为19.2%;根据调查的资料和获得的数据对其地理分布、种属、饲养管理等情况加以归纳、分析和总结。结论:无论是鸡还是猪,三种不同采样部位分离所得空肠弯曲杆菌的感染阳性率呈显著差异,其中盲肠内容物中空肠弯曲杆菌的感染阳性率最高,肝脏最低;就肝脏样品空肠弯曲杆菌检测结果来看,猪和鸡的带菌状况无显著差异,盲肠内容物空肠弯曲杆菌检测结果来看,猪的感染阳性率显著高于鸡;上海市内(奉贤等地)来源的畜禽感染阳性率显著低于浙江金华、江苏海门、江苏张家港和江苏大丰等外省市地区。该结论为掌握空肠弯曲杆菌的流行规律、预测其流行趋势、有效预防、诊断控制空肠弯曲杆菌病提供了一定的理论依据。
【Abstract】 Our study was concentrated on the epidemiological investigation of C.jejuni which was carried out to the chicken and swine from twenty-eight habit(Henan Kaifeng、Shandong Qingdao、Jiangsu Nantong、Zhejiang Jinhua、Shanghai Fengxian Etc) to Shanghai. 6702 samples (3657from swine and 3045 from chicken) including liver samples、appendix content samples、dejecta samples were collected according to principle——geometric proportion and random sampling. Quick sensitive and specific PCR was conducted when VS1 gene was selected as the target sequence which showed unique specificity to C.jejuni while other bacterium can’t amplify specific fragment with VS1 gene.The result of investigation and research indicated that there existed C.jejuni infected both in swine and chickens and different habit and different genera had different infection rate. 3045 samples of chickens(1015 samples of liver, appendix content and cloaca swab respectively) were detected by PCR. The result showed that infection positive rate of liver, appendix content and cloaca swab were 13.2%, 28.4%, 19.8% respectively. 3657 samples of swine(1219 samples of liver, appendix content and dejecta respectively) were detected by PCR. The result showed that infection positive rate of liver, appendix content and dejecta were 10.3%, 34.4%, 19.2% respectively. Geographic distribution genera and breeding management condition were induced, analyzed and summarized according to the data that we got in the investigation.Conclusion: no matter it was swine or chicken, there was significant deviation in C.jejuni infection positive rate of three different sampling position and sample of appendix content showed the highest infection positive rate while that of liver the lowest. There was no significant deviation in C.jejuni infection positive rate between Swine and chicken’s liver samples, while appendix content samples of swine was significant higher in C.jejuni infection positive rate than that of chicken. Swine and chicken from Shanghai showed significant lower C.jejuni infection positive rate than that from Zhejiang Jinhua, Jiangsu Haimen, Jiangsu Zhangjiagang Jiangsu Dafeng and other zones. The conclusion provide theory foundation to the handling of C.jejuni epidemic rules, anticipation of C.jejuni epidemic tendency and efficient preservation and diagnosis of C.jejuni diseases.
