新型双核大环多胺金属配合物的合成与DNA的性质研究

【摘要】 化学核酸酶是当今化学生物学中发展迅猛、富有挑战性的新领域之一。近20年来,人工核酸切割试剂尤其是定点切割试剂的研究一直是生物化学和分子生物学等学科中最为活跃的前沿领域之一。大环多胺类金属配合物广泛应用于生物、化学和医学领域。本文致力于新型双核大环多胺金属配合物的设计合成与仿酶催化性能的研究。本文以大环为原料,合成了以二肽桥联的双核大环多胺及其过渡金属配合物,其结构通过了1~HNMR、质谱、红外以及元素分析等方法的鉴定,研究了目标化合物的合成方法,实现了简便高效的选择性合成。研究了此类配合物作为化学核酸酶模型对质粒DNA(pUC19)的催化裂解性能,结果表明:配合物分子在生理条件下,对质粒DNA(pUC19)有很好的切割活性,成功模拟了化学核酸酶切割DNA的高效性和专一性。
【Abstract】 Artificial nucleases are a new challenging filed that is quickly developing in chemical biology. In this 20 years, the study that chemical nucleases reagents especially selectivity site cleavage reagents is the most active field in chemical biology.Macroyclic polyamines metal complexes as enzyme models are widely used in chemical biology, medicine and gene therapy. In this thesis, novel Macroyclic polyamines and their metal complexes have been designed and catalytic efficiency in DNA cleavage.A series of multinuclear macrocyclic polyamine metal (Zn2+ Cu2+ Co2+) complexes containing chiral dipeptide linkage were designed and synthesized. The structures of these novel dinuclear complexes were consistent with the date obtained from HNMR, MS and IR et al. The synthetic methods of the title compounds have been discussion in detail, a series of convenient and efficient reactions have been developed. The hydrolysis cleavage of plasmid DNA (pUC19) by macrocyclic polyamine metal complexes has been investigated, and the results revealed that these complexes could act as powerful catalysts for the cleavage of Plasmid DNA under physiological conditions.
