川芎嗪对氧化损伤所致白内障抑制作用的实验研究

【摘要】 [目的]观察中药川芎嗪对过氧化氢诱导大鼠白内障的抑制作用。[方法]取SD大鼠24只,实验前散瞳观察晶状体情况,颈椎脱臼处死后立即摘取眼球,无菌及无损伤条件下取出晶状体并预孵3小时后,将透明晶状体分为5组,即空白对照组、过氧化氢组(过氧化氢终浓度为300μmol/L)、3个不同浓度川芎嗪组(实验干预组)。孵育24小时后取出晶状体,于空白对照组、过氧化氢组、川芎嗪Ⅰ组三组各取一晶状体进行扫描电镜观察分析,剩余晶状体观察晶状体混浊情况并拍照,观察各组晶状体白内障形成情况,然后予以匀浆后取上清液检测超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)、过氧化氢酶(CAT)的活性。[结果]观察各组晶状体可见:空白对照组晶状体孵育24小时后均保持透明,未见白内障形成;过氧化氢组晶状体孵育24小时后全部混浊,形成皮质性白内障;不同浓度川芎嗪组的晶状体混浊程度不一,药物浓度越高越透明,白内障形成愈不明显。晶状体混浊程度评分组间比较差异有统计学意义(F=9.2,P<0.05)。扫描电镜下见:空白对照组晶状体组织结构正常,过氧化氢组晶状体明显混浊,严重损害,川芎嗪Ⅰ组损害程度轻微。生化分析表明:与空白对照组比较,过氧化氢组SOD、GSH-PX、CAT活性显著降低(P <0.01),而川芎嗪保护组则有不同程度改善(P <0.01)。[结论]川芎嗪能增强晶状体中SOD、GSH-PX、CAT的活性,对大鼠过氧化氢所致白内障有抗氧化作用,从而延缓早期白内障的发生和发展,为临床白内障的诊疗提供了一定的思路。
【Abstract】 Objective To observe the inhibition of tetramethylpyrazine on oxidative-induced cataractous lens of rats.Materials and method: Twenty-four rats were killed and taken out of transparent lens with no injury. These lens were hatched in RPMI-1640 culture fluid for 3 hrs, then 33 transparent lens divided into 5 groups: normal control group, model group and three tetramethylpyrazine groups with different concentrations. The concentration of peroxide (H2O2) is 300μmol/L. The lens were hatched under the different conditions for 24 hrs in vitro. Three lens were examined by scanning electron microscopy. Then the transparency of the lens was observed. At the same time, the enzyme activation levels of Superoxide dismutase (SOD)、Glutathione peroxidase (GSH-PX) and Catalase (CAT) were measured respectively.Result: All lens were transparent in control group, all the lens of the model group developed cataract, the lens opacity of the three protective groups with tetramethylpyrazine were not so obvious as these of the model group lens by oxidative damage. The structure of len in normal control group was normal under the scanning electron microscopy but was damaged severely in modle group, slight damages were found in the len which were protected by tetramethylpyrazine. The difference of the score of the opacity between the groups is obvious(F=9.2,P<0.05).The enzyme activation levels of SOD、GSH-PX and CAT in the model group were obviously lower than those of enzymes in normal control group(P <0.01), but the levels of them increased in tetramethylpyrazine groups compared with those in model group(P<0.01).Conclusion Tetramethylpyrazine could protect lens of rats from oxidative damage by increasing enzyme activation of SOD、GSH-PX、CAT and inhibit or delay the generation and development of peroxidate-induced cataract at the early stage.
