应用LsAB技术显示五羟色胺受体亚型1A在小鼠螺旋神经元的分布

【摘要】 目的五羟色胺(serotonin,5-HT)是中枢神经系统中一种常见的神经递质,它通过与其独特的五羟色胺受体亚型相结合,对神经兴奋有着重要的调节作用。到目前为止,至少有七种五羟色胺受体系列被证实,且每个系列又包含有两种及两种以上的亚型,如五羟色胺1受体系列就分为A,B,D,E,F五种亚型,每个亚型与五羟色胺结合后通过与其匹配的不同的离子通道发挥不同的作用。其中有研究发现五羟色胺与神经组织的五羟色胺受体亚型1A结合后对该神经组织兴奋有着抑制调节作用。通过RT-PCR技术已证实小鼠螺旋神经元存在五羟色胺受体亚型1A,但到目前为止,从形态学方面对螺旋神经元上五羟色胺受体亚型1A的分布情况进行研究还未见报道。本实验旨在利用LsAB技术(酶标链亲和素-生物素技术)从形态学方面显示五羟色胺受体亚型1A在螺旋神经元上的分布。方法采用出生后三到六天的乳小鼠耳蜗进行冰冻切片,应用LsAB技术显示五羟色胺受体亚型1A在螺旋神经元上的分布情况。结果实验组的乳小鼠耳蜗螺旋神经元的细胞膜显示阳性信号,阴性对照组的乳小鼠耳蜗螺旋神经元的细胞膜上未发现阳性信号。结论五羟色胺受体亚型1A存在于乳小鼠耳蜗螺旋神经元的细胞膜上,推测当毛细胞刺激螺旋神经元引起它兴奋从而进行听觉信息传导时,五羟色胺作为神经递质与螺旋神经元的五羟色胺受体亚型1A结合可能对它的听觉传导活动有着抑制调节作用。

【Abstract】 Objective The serotonin is a widespread neurotransmitter in the centralnervous system (CNS), It plays an important role in modifying excitatory effectof nerves which is accomplished with the special serotonin receptor subtypesfrom the nerves. At present, at least seven different families of serotonin receptorshave been identified and each family of these receptors has two or more subtypes.For example the serotonin receptor 1 family consists of serotonin receptor 1A,serotonin receptor 1B, serotonin receptor 1D, serotonin receptor 1E, and serotoninreceptor 1F. Those functions are accomplished with the special serotonin receptorsubtypes that use different second messengers, which are coupled to distinct ionchannels. Some researches are presented that the serotonin can inhibit excitationof nerves when they combine with the serotonin receptors 1A which are locatedin the nerves. We know that the serotonin receptor 1A survives in the spiralganglion which was identified with RT-PCR technique, but we do not know itsspecific localization in the spiral ganglion. So in the present study, we locate theexpression of serotonin receptor 1A in the spiral ganglion with LsAB technique.Methods In the mouse(born between three to six days) cochlear frozen slides,serotonin receptor 1A in the spiral ganglion was located with LsAB technique(labeled streptavidin-biotin technique). Results Positive signals were seen inthe membrane of spiral ganglion As a negative control with no serotonin receptor1A antibody, no signal was found in the membrane of spiral ganglion. ConclusionThe serotonin receptor 1A has been found in the membrane of spiral ganglion. Itindicates that serotonin as a neurotransmitter maybe play an important role ininhibiting the auditory transmission of the spiral ganglion by the way that it combines with the serotonin receptor 1A which is located in the membrane of thespiral ganglion when it is stimulated by the hair cells to be excitatory.
